黑绛丹治疗^(60)Co射线照射致大鼠放射性皮炎的疗效评价 被引量:8

Evaluation of Heijiangdan Ointment on Radiodermatitis Induced by ^(60)Co γ-ray in Mice

摘要 目的探讨中药黑绛丹对大鼠Ⅳ度放射性皮炎的疗效及创面组织形态学和胶原变化的影响。方法 60Co射线6000cGy一次照射雌性Wistar大鼠,建立Ⅳ度放射性皮炎模型,将48只造模成功大鼠随机分为4组,每组12只。各组大鼠创面分别外敷黑绛丹、重组人表皮生长因子(rhEGF)外用溶液、Trolox治疗液、生理盐水,另设空白组12只,予生理盐水外敷创面,各组均每天治疗1次,连续治疗21天。分别于第1、6、11、16、21天测量各组大鼠创面面积,并在第11、21天创面组织取材并进行HE染色、维多利亚蓝+丽春红S双重组合染色和天狼猩红染色,观察创面组织形态学及胶原变化。结果第6、11、16、21天,黑绛丹组和rhEGF组大鼠创面面积均小于模型组(P<0.05);黑绛丹组创面面积小于Trolox组(P<0.05),但大于rhEGF组(P<0.05)。治疗21天后黑绛丹组大鼠创面组织炎症细胞少于模型组,弹力纤维、胶原纤维以及Ⅰ型和Ⅲ型胶原较模型组增多。结论黑绛丹能加速大鼠Ⅳ度放射性皮炎创面的愈合,减轻创面的炎症反应,促进弹力纤维和胶原的修复。 Objective To investigate the effect of Heijiangdan Ointment (Ointment of carbonized human hair and yolk) on the Ⅳ degree radiation dermatitis in mice and its influence on the histomorphological and collagenous changes of wound. Methods Forty-eight Wistar female mice with the Ⅳ degree radiation dermatitis induced by 60Co γ-ray 6 000cGy one time radiation were randomized into 4 groups with 12 in each. The 4 groups were treated with the application of Heijiangdan Ointment, recombinant human epidermal growth factor (rhEGF) solution, Trolox solution, and normal saline respectively. A blank group was set up with 12 mice in it and treated with application of normal saline. The treatment for all groups was once a day and 21 days in succession. On the 1st, 6th, 11th, 16th, and 21st day of treatment, the area of ulcer of all groups were measured, and on the 11th and 21st day, the ulcer tissues were collected and stained by HE staining, Victoria blue with Ponceau S staining, and Sirius red staining for observing the changes in histomorphology and collagen of the wound. Results On 6th,11th, 16th and 21th day, the area of skin ulcer in Heijiangdan Ointment group and rhEGF group was smaller than that in the model group (P0.05), the area of skin ulcer in Heijiangdan Ointment group was smaller than that in the Trolox group (P0.05) but bigger than that in the rhEGF group (P0.05). After the treatment for 21 days, the inflammatory cells in wound tissues in the Heijiangdan Ointment group was less than those in the model group, while the elastic fibers, collagenous fibers and collagens typeⅠand type Ⅲ were more than those in the model group. Conclusion Heijiangdan Ointment may speed up the ulcer healing of the Ⅳ degree radiation dermatitis, reduce the inflammatory reaction in wound, and promote the repair of elastic fibers and collagen.
