摘要
以海州常山带芽茎段为外植体,研究不同消毒方法、基本培养基种类、植物生长调节剂配方和活性炭对其离体培养及植株再生的影响,建立海州常山离体快繁技术体系。结果表明:海州常山带芽茎段的最佳消毒方法为75%酒精处理30 s,再用0.1%HgCl_2溶液处理7 min;芽萌发的最适培养基为MS+6-BA 0.20 mg/L+IAA 0.10 mg/L,萌发率可达86.67%;茎段最适增殖培养基为MS+6-BA 1.00 mg/L,增殖系数为4.47;最适生根培养基为1/2 MS+IBA 1.20 mg/L,生根率为86.67%,平均生根数6.02。本研究建立了海州常山茎段的再生体系,为良种繁育和推广应用提供了技术支持。
The effects of different disinfection methods, basic medium species, plant growth regulator formula and activated carbon on its in vitro culture and plant regeneration were taken as explants in Clerodendrum trichotomum bud stem segments, and the rapid propagation system of Clerodendrum trichotomum in vitro was established. The results showed that the most appropriate method of surface sterilization on stems of C. trichotomum was 75% alcohol for 30 seconds and 0.1% HgC12 for 7 minutes. The fittest medium for germination of stem axillary bud was MS+6-BA 0.20 mg/L+IAA 0.10 mg/L, with 86.67% germination rate ofaxillary bud. The best medium for shoot multiplication was MS+6-BA 1.00 mg/L, and multiplication on coefficient could reach 4.47. The best rooting medium was 1/2 MS+IBA 1.20 mg/L, with 86.67% rooting rate and average rooting number of 6.02. This study established the regeneration system of stem segments of C. trichotomum, which would provide the technical support for seedling production and promotion.
