摘要
目的建立紫杉醇(PTX)-PEG功能化多壁碳纳米管(DSPE-PEG-MWCNTs-PTX)包封率的测定方法。方法采用葡聚糖G-50凝胶微柱离心法分离DSPE-PEG-MWCNTs与游离药物,用HPLC法测定结合在DSPE-PEG-MWCNTs的PTX,计算包封率。结果在本法选择的色谱条件下,PTX得到良好的分离,在2~200μg/m L(r=0.999 7)线性关系良好。葡聚糖G-50凝胶微柱离心法能有效分离DSPE-PEGMWCNTs与游离药物,测得DSPE-PEG-MWCNTs-PTX平均包封率为88. 17%,RSD为2. 33%(n=3)。结论建立的葡聚糖凝胶微柱离心-HPLC法简便快捷,测定结果准确可靠,可用于测定DSPE-PEG-MWCNTs-PTX包封率。
Objective To establish a method for determining the entrapment efficiency of paclitaxel-PEG functionalized multi-carbon nanotubes(DSPE-PEG-MWCNTs-PTX). Methods Sephadex G-50 microcolumn centrifugation was used to separate free drug from DSPE-PEG-MWCNTs. The PTX bound to DSPE-PEG-MWCNTs was quantified by HPLC. Results Sephadex G-50 microcolumn centrifugation could separate DSPE-PEG-MWCNTs and free drug,and the linear range of PTX was 2~200 μg/mL ( r =0.999 7).The average entrapment efficiency of DSPE-PEG-MWCNTs-PTX was 88.17% with RSD 2.33% ( n =3). Conclusion The developed microcolumn centrifugation-HPLC method is simple,rapid,accurate and reliable for the determination of the entrapment efficiency of DSPE-PEG-MWCNTs-PTX.
