摘要 目的:优选白屈菜粗多糖的水提醇沉工艺。方法:以白屈菜多糖得率和多糖含量为指标,以水为溶剂,选取料液比、提取时间、提取温度、提取次数、醇沉浓度为影响因素,进行单因素考察,在综合单因素实验的基础上,采用L9(3~4)正交试验,以加权评分法综合评估白屈菜多糖提取条件,对白屈菜多糖水提工艺进行优化,采用苯酚-硫酸法测定多糖的含量。结果:白屈菜多糖的最佳提取工艺为30倍量的水,100℃提取3次,每次3h,滤液浓缩至料液比1∶4,加入95%乙醇至含醇量达到80%,静置过夜,离心,沉淀,依次用无水乙醇、丙酮、无水乙醚洗涤,挥干乙醚即得。结论:此工艺操作简便、成本低廉、重现性好、适合大规模工业生产,可用于白屈菜药材及饮片中多糖的提取。 Objective: To optimize water extraction and alcohol precipitation process of polysaccharide from Chelidonium majus. Methods: On the basis of single factor test with materia liquid ratio, extraction time, extract temperature, extraction times and alcohol concentration of alcohol-precipitation procedure for influence factors. On the basis of single factor experiment,the orthogonal test with 4 factors and 3 levels method was used to design and the content of the extract was determined using anthrone-sulfuric acid colorimetry method. Results: The best extracting process was carried out by water(1 : 30), extracting 3 times under 100℃, each time 3 hours. The filtrate is concentrated to the mixture ratio of 1 :4 and the 95% ethanol was added to the alcohol content reaches 80%, rest over night, centrifuge, the precipitation in turn is washed with anhydrous ethanol, acetone, anhydrous ether, with the drying of ether. Conclusion:This process is simple, inexpensive, reproducible and suitable for large-scale industrial production, It can be used for the extraction of polysaccharides from Chelidonium majus.
