摘要
目的评估一种高特异性沙门菌显色培养基(HS)的性能。方法沙门菌、其他菌及实际样品按GB 4789.4—2010接种到HS沙门菌显色培养基、CHROMagar显色培养基及XLD传统培养基上,培养后观察结果。结果 35株沙门菌在HS、CHROMagar和XLD上特异性分别为100.00%、88.57%和100.00%。54株非沙门菌在HS、CHROMagar和XLD上假阳性率分别为1.85%、20.37%和77.78%。48份样品中沙门菌3种培养基确认阳性率均为2.08%,其中HS、CHROMagar和XLD符合率分别为100.00%、16.67%和20.00%。结论纯菌及实际样品检测结果表明,HS高特异性显色培养基上沙门菌假阴性、非目标菌假阳性率最低,特异性最好,食品基质中沙门菌的检出准确度也是HS最好。HS对提高食品中沙门菌检验的效率有重要意义。
Objective To evaluate the performance of Salmonella chromogenic medium with high specificity. Methods A total of 35 strains of Salmonella spp,54 other strains and food samples were inoculated onto HS,CHROMagar chromogenic media and XLD conventional isolation medium for observation according to GB 4789. 4 2010. Results The specificity of the 35 Salmonella species on HS,CHROMagar and XLD was 100. 00%,88. 57% and 100. 00% respectively. The false-positive rate of the 54 non-Salmonella on HS,CHROMagar and XLD was 1. 85%,20. 37% and 77. 78%,respectively. The total positive detection rate of Salmonella on HS,CHROMagar & XLD all was 2. 08% in 48 samples. The coincidence rate of HS,CHROMagar and XLD was 100. 00%,16. 67% and 20. 00%,respectively. Conclusion Pure culture test results showed that HS has the lowest false-positve ratio. Food matrix isolation and identification results also indicated that HS was more specific than the other chromogenic media and conventional media. It has the best accuracy in isolation and presumptive identification of Salmonella. The HS medium offers a high degree of specificity for the detection of Salmonella in food samples.
