摘要
目的比较荧光PCR法与细菌培养法对从业人员肠道携带沙门菌和志贺菌的检出率。方法收集2015年7月24日-2015年11月13日4 620份从业人员肛拭子,分别用荧光PCR法和培养法进行肠道沙门菌和志贺菌的检测,比较2种方法的检出率。结果培养法检出沙门菌54株、志贺菌0株,总检出率为1.17%;荧光PCR法检出沙门菌81株、志贺菌1株,总检出率为1.78%。荧光PCR法检测的灵敏度高于培养法检测,差异有统计学意义(χ~2=5.85,P〈0.05)。荧光PCR法与培养法对比,其灵敏度为100.00%,特异度为99.39%。结论荧光PCR方法操作简便、快速、灵敏度高、特异性强,可降低工作强度,适合在实验室进行常规推广;本区从业人员肠道沙门菌携带率明显偏高,相关部门需加强公共卫生监督。
Objective Making a comparison between the detection rates of Salmonella and Shigella in occupational personnel’intestinal tracts using fluorescence PCR method and bacterial culture method respectively. Methods Fluorescence PCR method and culture method were used respectively to test Salmonella and Shigella in intestinal tracts,which were taken from 4 620 anal swabs of practitioners from July 24,2015 to November 13,2015,and then the detection rates of the two methods were compared. Results 54 strains of Salmonella and 0 Shigella were detected by culture method,with the total positive rate of1. 17%; 81 strains of Salmonella and 1 strain of Shigella were detected by fluorescence PCR method,with the total positive rate of 1. 78%; the sensitivity of fluorescent PCR detection method was higher than culture method,and the difference was statistically significant( χ~2= 5. 85,P 〈 0. 05). Compared with culture method,the sensitivity was 100. 00% and the specificity was99. 39% with fluorescence PCR method. Conclusion Fluorescence PCR method has the advantages of simple operation,rapidness,high sensitivity and strong specificity,which can reduce the work intensity,being suitable for regular promotion in laboratory. The carrying rate of enteric Salmonella of practitioners in our district was obviously higher,which needs relevant departments to strengthen public health supervision.
