电针长强穴对自闭症大鼠海马CA3区BDNF蛋白表达的影响 被引量:4

Effect of Electro-acupuncture at Changqiang Acupoint on the Expression of BDNF Protein in Hippocampus CA3 of Rats with Autism

摘要 目的:观察电针长强穴对自闭症大鼠海马CA3区脑源性神经营养因子(BDNF)蛋白表达的影响。方法:采用孕鼠腹腔注射丙戊酸钠的方法造模,取自闭症大鼠24只,采用随机数字表法分为模型组、长强组和非穴组,每组8只;另取正常生产的仔鼠8只,作为空白组。长强组采用0.5寸自制的双极联体针直刺长强穴0.3寸,予电针干预20 min,20 d/疗程;非穴组除取穴外,余操作同长强组;空白组和模型组同等条件饲养。在大鼠出生后7 d(PN7)、PN14、PN23和PN34对各组大鼠进行称重,采用Morris水迷宫观察治疗后大鼠学习记忆能力的变化,应用免疫组化技术观察各组大鼠海马CA3区BDNF蛋白的表达。结果:(1)体质量:在PN34时,长强组的体质量水平高于模型组(P<0.05);非穴组与模型组比较,差异无统计学差异(P>0.05)。(2)Morris水迷宫:与空白组相比,模型组平均潜伏时间明显延长,原平台所在象限游泳时间/总时间明显减少,差异均有统计学意义(P<0.05);与模型组相比,长强组平均潜伏时间明显缩短、原平台所在象限游泳时间/总时间明显增多,差异均有统计学意义(P<0.05),非穴组平均潜伏时间缩短,原平台所在象限游泳时间/总时间增多,差异均无统计学意义(P>0.05)。(3)免疫组化:与空白组相比,模型组BDNF蛋白平均灰度值表达明显升高,差异有统计学意义(P<0.05);与模型组相比,长强组BDNF蛋白平均灰度值表达明显降低,差异有统计学意义(P<0.05),非穴组BDNF蛋白平均灰度值表达降低,差异无统计学意义(P>0.05)。结论:电针长强穴可提高自闭症大鼠海马CA3区BDNF蛋白的表达,从而改善自闭症大鼠的生长发育和学习记忆能力。 Objective: To explore the effects of electro-acupuncture at Changqiang acupoint on the BDNF protein expression in hippocampal CA3 region in rats with autism. Methods: Autism rat model was made by intraperitoneal injection of pregnant rats with valproate sodium(VPA). A total of 24 autism rats were divided into model group, Changqiang group and non-acupoint group, with8 rats in each group; 8 normal litter mate rat were used as blank group. The Changqiang group acupuncture Changqiang point 0.3 inch,using 0.5 inch homemade bipolar conjoined needle, electro-acupuncture 20 min, 20 days a course of treatment; The non-acupoint group adopts the same method as the Changqiang group except the acupoints are different; The blank group and model group were kept under the same condition. Rats were weighed at different time points after birth(PN7, PN14, PN23 and PN34), using Morris water maze to evaluate learning and memory ability of rats, and using immunohistochemical to evaluate BDNF protein expression in hippocampal CA3 area of rats. Results: 1. Weight: At PN34, weight of Changqiang group was higher than the model group(P〈0.05);there was no statistical difference between non-acupoint group and model group(P〉0.05). 2. Morris sater maze: Compared with the blank group, average latency time of the model group was significantly prolonged, quadrant swimming time/total time of the original platform decreased significantly, with statistically significant difference(P〈0.05); Compared with the model group, average latency time was significantly shortened, quadrant swimming time/total time of the platform increased significantly, with statistically significant difference(P〈0.05), the average latency time of the non-acupoint group was significantly shortened, quadrant swimming time/total time of the platform increased significantly, with no statistically significant difference(P〉0.05). 3.
