基于轮廓分析的电针命门穴对FMR1基因敲除小鼠不同脑区CREB蛋白表达的影响 被引量:2

Electroacupuncture at Mingmen Point Based on Profile Analysis has Effect on CREB Expression in Different Brain Regions of FMR1 Gene Knockout Mice

摘要 目的:观察电针命门穴对FMR1基因敲除小鼠不同脑区环磷腺苷效应元件结合蛋白(CREB)及p-CREB表达的影响,以探讨其空间分布特征。方法:采用随机数字表法将适龄FMR1基因敲除小鼠分为空白组、非经非穴组和命门组,每组8只。空白组仅模拟抓取动作。命门组选取小鼠命门穴,非经非穴组选取小鼠后海穴旁开约1 cm的部位,每天固定时间电针干预,频率2 Hz,强度2 mA,连续波,每天30 min,连续干预14 d。采用Western blot法检测小鼠小脑、皮质及海马区CREB及p-CREB蛋白表达。结果:与空白组比较,命门组及非经非穴组海马区CREB蛋白表达升高(P<0.05)。与同组皮质区比较,各组小鼠小脑及海马区CREB蛋白表达均升高(P<0.05)。不同脑区间CREB蛋白表达趋势相同,趋势变化程度不同。与空白组比较,非经非穴组小脑及皮质区p-CREB蛋白表达降低(P<0.05)。与同组小脑比较,非经非穴组海马区p-CREB蛋白表达升高(P<0.05);与同组皮质区比较,命门组、非经非穴组海马区p-CREB蛋白表达升高(P<0.05),不同脑区间p-CREB蛋白表达趋势不同。与非经非穴组比较,命门组各指标差异无统计学意义。结论:电针命门穴在不同脑区中能相应地强化FMR1基因敲除小鼠海马区CREB的磷酸化,促进大脑功能的重组和代偿。 Objective:To observe the effect of electroacupuncture at Mingmen point(GV4)on expressions of cAMPresponse element-binding protein(CREB)and p-CREB in different brain regions of FMR1 gene knockout mice,so as to explore its characteristics of spatial distribution.Methods:The FMR1 gene knockout mice of the right age were divided into the blank group,the non-meridian-non-point group and the Mingmen group according to the random number table method,8 mice in each group.The blank group was only given the simulation of grasping.The Mingmen points of mice in the Mingmen group and the sites being 1 cm near the Houhai point of mice in the non-meridian-non-point group were selected,both of which received electroacupuncture intervention at a fixed time for 30 minutes for 14 consecutive days,with a frequency of 2 Hz,an intensity of 2 mA and a continuous wave.The expression of CREB and p-CREB in cerebellum,cortical area and hippocampus of mice was detected by Western blot.Results:Compared with that in the blank group,CREB expression in hippocampus of mice in the Mingmen group and the non-meridian-non-point group was increased(P<0.05).Compared with that in the cortical area in the same group,CREB expression in cerebellum and hippocampus in each groups was increased(P<0.05).The trend of CREB expression in different brain regions was the same,but the variation degree was different.Compared with that in the blank group,p-CREB expression in cerebellum and cortical area in the non-meridiannon-point group was decreased(P<0.05).Compared with that in cerebellum in the same group,p-CREB expression in hippocampus in the non-meridian-non-point group was increased(P<0.05).Compared with that in the cortical area in the same group,p-CREB expression in hippocampus in the Mingmen group and the non-meridian-non-point group was increased(P<0.05),and the trends of p-CREB expression in different brain regions were different.Compared with those in the non-meridian-non-point group,there was no significant difference being found in each index in the M
