摘要
目的:本研究建立P388/VCR细胞白血病多药耐药模型,予小、中、大剂量的扶正祛邪复方中药联合长春新碱(VCR)作用于裸鼠模型,检测B淋巴细胞瘤-2基因(Bcl-2)的表达。方法:建立裸鼠P388/VCR细胞耐药模型,随机分成正常(NOR)、模型(MOD)、化疗(VCR)、中药小剂量+VCR(LOW)、中药中剂量+VCR(MED)、中药大剂量+VCR(HIG)组。观察用药前、后裸鼠的精神、觅食、皮肤色泽、活动度,测量用药后裸鼠瘤体重量情况,计算抑瘤率。切除瘤体后用RT-PCR检测Bcl-2表达量。结果:VCR、LOW、MED、HIG抑瘤率分别是:30.66%,39.02%,44.25%,55.4%。中药各组抑瘤率均大于化疗组。Bcl-2表达情况:与VCR组相比,中药各组Bcl-2表达量均出现显著降低(P<0.05);各中药浓度组之间比较,HIG组Bcl-2表达量高于MED组和LOW组(P>0.05)。说明扶正祛邪复方中药逆转P388/VCR细胞耐药机制可能是通过下调Bcl-2表达实现的,与中药浓度无关系。结论:(1)扶正祛邪复方中药能抑制P388/VCR细胞增长;(2)扶正祛邪复方中药可降低P388/VCR裸鼠模型Bcl-2的表达,与中药浓度无关;(3)逆转白血病耐药细胞株P388/VCR的机制之一可能与下调Bcl-2的表达相关。
Objective： To establish a model of P388/VCR cell leukemia with multidrug resistance. Reversing the multidrug resistance of leukemia with small,medium and high doses of medicine with function of supporting right and dispelling devil compound combined with vincristine（ VCR）,and to detect the expression of B Lymphoblastoma-2 gene（ Bcl-2）. Method： Drug resistance model of P388/VCR cells in nude mice was established and randomly assigned to NOR,MOD,VCR,LOW,MED,and HIG. Observed the spirit,foraging,skin color and activity of nude mice before and after treatment,and the weight change of the nude mice after the treatment. The tumor inhibition rate was calculated. Bcl-2 expression was detected by RT-PRC after resection of tumor. Results： The tumor inhibition rate of VCR,LOW,MED,HIG were： 30. 66%,39. 02%,44.25%,55. 4% respectively. The tumor inhibition rate of TCM group was higher than that of chemotherapy group.Compared with the VCR group,the expression of BC1-2 in every TCM group reduced significantly（ P 0.05）. Comparison between the concentration groups of traditional Chinese medicine showed the expression of BCl-2 in HIG group was higher than that in MED group and LOW group（ P〉0. 05）,which showed the mechanism of reversing drug resistance of P388/VCR cells by TCM with the function of supporting right and dispelling devil may be achieved by down-regulating the expression of Bcl-2,which has no relationship with the concentration of TCM. Conclusion：（ 1） The growth of the tumor cells could be inhibited by TCM with the function of supporting right and dispelling devil.（ 2） The expression of Bcl-2 in P388/VCR nude mice can be reduced by TCM with the function of supporting right and dispelling devil,and it has no relationship with the concentration of TCM.（ 3） One of the mechanisms of reversing P388/VCR may be related to down-regulation of Bcl-2 expression.
