Effect of atrial natriuretic peptide on Thl7 cell differentiation in bronchiolitis mice

摘要 的 研究心房利钠肽(atrial natriuretic peptide,ANP)对毛细支气管炎(简称毛支)模型小鼠肺组织辅助性T细胞(helper T cell,Th)17细胞分化的影响,为毛支治疗提供理论基础.方法 按照随机对照原则将ALB/C小鼠分为正常组、呼吸道合胞病毒(respiratory syncytial virus,RSV)组、ANP+ANP抑制剂(A71915)组和ANP组,滴鼻法成功建立毛支模型,而后雾化吸入ANP或腹腔注射A71915,收集支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)检测白细胞介素(interleukin,IL)-17含量,留取肺组织进行HE染色、免疫组织化学染色和Western blot检测,观察RORγt的蛋白表达及气道炎症情况.结果 与正常组相比,RSV组小鼠IL-17及RORγt的含量增高(P〈0.05);与RSV组相比,ANP组小鼠IL-17及RORγt的表达量均降低(P〈0.05),ANP+A71915组各项指标较RSV组相比无显著差异(P〉0.05).结论 ANP能够抑制毛支小鼠Th17细胞的分化.  Objective To investigate the effects of eatrial natriuretic petide(ANP)on the differentia-tion of Th17 cells in the lung tissue of bronchiolitis mice and provide a theoretical basis for the treatment of bronchiolitis. Methods BALB/ C mice were divided into normal group,bronchiolitis group(RSV),ANP +A71915 group and ANP group,The bronchiolitis model was successfully established by intranasal nasal drip-ping,and then treated with ANP or A71915 to collect bronchoalveolar lavage fluid(BALF)to detect the level of IL-17,the lung tissue was taken for RORγt and airway inflammation by HE staining,immunohistochemical stai-ning and Western blot. Results Compared with the normal group,the levels of IL-17 and RORγt in the RSV group were significantly increased. Compared with the RSV group,the expression of them in the ANP group was significantly decreased (P 〈 0. 05). There was no significant difference between ANP + A 71915 group and RSV group(P 〉 0. 05). Conclusion ANP can inhibit the differentiation of Th17 cells in bronchiolitis mice.
