摘要
【目的】探讨补肾活血化痰开窍方对耳鸣模型大鼠肾虚相关指标的影响。【方法】将60只大鼠随机分为正常组,模型组,中药低、中、高剂量组,西药组,每组10只。除正常组外,其余组别大鼠采用水杨酸钠腹腔注射联合"饮水抑制"大鼠行为学实验复制耳鸣模型。造模成功后,正常组、模型组大鼠给予2 m L生理盐水灌胃;中药低、中、高剂量组给予补肾活血化痰开窍方(剂量分别为5.5、11、22 g·kg^(-1)·d·^(-1))灌胃;西药组大鼠给予卡马西平(剂量为5 mg·kg^(-1)·d·^(-1))灌胃。每天1次,持续给药8周。采用全自动生化仪测量大鼠血清钙、磷、铁,采用酶联免疫吸附法(ELISA)测定大鼠血浆多巴胺、皮质醇、环磷酸腺苷(c AMP)、环磷酸鸟苷(cGMP)。【结果】造模成功率为88%。与正常组比较,模型组大鼠血液中钙、铁、皮质醇、多巴胺、cAMP含量,cAMP/cGMP比值下降,磷、cGMP含量增加(均P<0.05);与模型组比较,中药各剂量组上述指标明显改善(P<0.05)。【结论】补肾活血化痰开窍方可改善耳鸣大鼠肾虚相关指标。
Objective To investigate the effects of Bushen Huoxue Huatan Kaiqiao Recipe（BHHKR;a recipe with the actions of tonifying kidney,activating blood,resolving phleym for resuscitation）on kidney deficiency-related blood indexes in rat model of tinnitus. Methods The 60 rats were randomly divided into normal group, model group,low-,middle-and high-dose Chinese medicine groups,western medicine group,10 rats in each group.Except for the normal group, the rats in the other groups were induced into tinnitus model by intraperitoneal administration of sodium salicylate combined with water deprivation test. After successful modeling,the normal group and the model group were given intragastric administration of 2 mL of normal saline,the low-,middle-and high-dose Chinese medicine groups were given intragastric administration of BHHKR at the dosage of 5.5,11,22 g·kg^-1·d·^-1,respectively,western medicine group was given intragastric administration of carbamazepine at the dosage of 5 g·kg^-1·d·^-1, one time per day,the treatment lasting for 8 weeks. After treatment,the serum calcium,phosphorus and iron were measured by automatic biochemical analyzer,and plasma dopamine,cortisol,cAMP and cGMP were measured by enzyme-linked immunosorbent assay（ELISA）. Results The successful modeling rate was 88%. Compared with the normal group,in the model group,the contents of blood calcium,iron,dopamine,cortisol,c AMP and cAMP/cGMP ratio were decreased,and the contents of phosphorus and cGMP were increased（P〈0.05）. Compared with the model group,the above indexes in various Chinese medicine groups were improved（P〈0.05）. Conclusion BHHKR has effects on kidney deficiency-related blood indexes in rat model of tinnitus.
