一种基于荧光定量PCR的自动化检测装备初探 被引量:2

Preliminary Study of Automatic Inspection Equipment Based on Quantitative PCR Method

摘要 试验以食药品中必检微生物沙门氏菌为供试菌株,针对实验室首次自主研发的微生物分子检测仪,建立一种配套该仪器使用的沙门氏菌荧光定量PCR自动化检测体系,该微生物分子检测仪可以同时完成核酸提纯和荧光定量检测。在该设备调试成功后,对某高校食堂14个食品样本进行自动化检测。结果表明,该q PCR体系的准确性好、特异性强;相比于其他检测方法,该体系的检测下限更低且时效性更佳;经过与沙门氏菌检测的国家标准比较,建立的配套设备的检测体系可以快速、自动化、高通量地检测不同食品基质中沙门氏菌的数量,且结果可靠。 Adopt the Salmonella as the tested strains which was required inspection item in the food and drug. A quantitative PCR detection system for Salmonella was established to support a microbial molecular detector which was made independently in laboratory at the first time. The microbial molecular detector can complete nucleic acid purification and fluorescent quantitative detection at the same time. The food samples from a canteen in college were detected automatically after the device has been debugged successfully. The result showed that the accuracy and specificity were good, and there were lower detection limit and better timeliness in the q PCR detection system. The system could rapidly detect the quantity of Salmonella from the most food samples and the result was reliable which compared with national standards of Salmonella detection.
