摘要
目的探讨姜黄素联合阿霉素对肝癌PLC/PRF/5细胞增殖、凋亡及化疗药物增敏的作用,及其可能的作用机制。方法根据不同的干预药物将细胞分为3组,对照组、阿霉素组、姜黄素联合阿霉素组(联合组),MTT法检测3组肝癌PLC/PRF/5细胞增殖抑制率,RT-PCR检测肿瘤细胞Caspase-3基因的表达,TUNEL免疫荧光检测肿瘤组织凋亡细胞数,比较裸鼠进行不同药物治疗前后肿瘤组织的体积及治疗前后的体积差。结果阿霉素组细胞抑制率高于对照组(P <0. 05),联合组细胞抑制率高于阿霉素组(P <0. 05);阿霉素组Caspase-3基因表达高于对照组(P <0. 01),联合组Caspase-3基因表达高于阿霉素组(P <0. 01);在相同视野范围内,阿霉素组凋亡的肿瘤细胞数多于对照组(P <0. 05),联合组凋亡的肿瘤细胞数多于阿霉素组(P <0. 05)。三组裸鼠治疗前肿瘤体积比较差异无统计学意义(P> 0. 05),阿霉素组治疗后肿瘤体积及治疗前后体积差均小于对照组(P<0. 01),联合组肿瘤体积及治疗前后体积差均小于阿霉素组(P <0. 01)。结论姜黄素与阿霉素联合使用较单独使用阿霉素能增加PLC/PRF/5细胞的药物敏感性,这种作用可能与二者联用后能有效抑制PLC/PRF/5细胞增殖、促进PLC/PRF/5细胞的凋亡有关。
Objective To investigate the effects o f curcumin combined with adriamycin on the proliferation, apoptosis and the sensitization of chemotherapeutics in PLC/PRF/5 cells of liver cancer and its possible mechanism. Methods PLC/PRR/5 cells were divided into control group, adriamycin group and curcumin combined with adriamy-cin group (combination group). MTT assay was used to detect the survival rate of PLC/PRF/5 cells. RT-PCR was used to detect the Caspase-3 gene expression. The number of apoptotic cells in tumor tissue was detected by TUNEL immunofluorescence. The volume and its difference value of tumor tissue before and after different drug treatment were compared. Results The inhibitory rate of cells in adriamycin group was higher than that of control group ( P 〈0. 0 5 ) , and combination group was higher than adriamycin group ( P 〈 0.05). The Caspase-3 gene expression in adriamycin group was higher than that in control group ( P 〈0. 01) ,and combination group was higher than adriamycin group ( P 〈0. 01). In the same field of view,the number of apoptotic tumor cells in adriamycin group was more than that in con-trol group (P 〈0.05), and combination group was more than adriamycin group ( P 〈 0.05). The tumor volume of nude mice in each group before treatment was not significantly different ( P 〉0.05) , but after treatment, the tumor vol-ume of adriamycin group and its difference value were lower than those of control group ( P 〈0. 01) ,and those in com-bination group was the lowest ( P 〈 0. 01). Conclusion Curcumin combined with adriamycin can increase the drug sensitivity of PLC /PRF /5 cells compared with adriamycin alone. This effect may be related to inhibiting the prolifera-tion and promoting the apoptosis of PLC/PRF/5 cells after combination use.
