摘要
目的:观察中药黄连浸提液对耐甲氧西林金黄色葡萄球菌(MRSA)、产广谱β-内酰胺酶(ESBL)肺炎链球菌(S.pneumoniae)、耐亚胺培南铜绿假单胞菌(P.aeruginosa)、产ESBL大肠埃希菌(E.coli)等4种常见临床耐药菌株的体外抑菌活性分析。方法:选取中药黄连制备水浸提液,依据CLSI标准,初步采用纸片琼脂扩散法(K-B法)测算黄连浸提液对MRSA、产ESBL的E.coli和S.peneumoniae、耐亚胺培南P.aeruginosa等菌株的抑菌直径并参考相应的标准菌株实验结果,再采用肉汤稀释法观察黄连浸提液对其相应抑菌活性,测定其相应的最小抑菌浓度(MIC)。结果:黄连浸提液对S.peneumoniae、MRSA、E.coli和P.aeruginosa等耐药菌株的抑菌直径分别为21,17,15,10 mm;MIC分别为15.63,62.50,125,250 mg·ml^(-1)。结论:黄连水浸提液对4种临床常见的耐药菌都有较好的抑菌效果,其中产ESBL的S.peneumoniae的抑菌效果最为明显,MRSA效果次之,其次为产ESBL的E.coli和耐亚胺培南P.Aeruginosa。本研究结果对肺炎链球菌及MRSA感染治疗具有重要的参考价值。
Objective: To observe the antibacterial activity in vitro of Coptidis Rhizoma on four common clinical drug-resistant strains, methicillin resistant Staphylococcus aureus, ESBL producing Streptococcus pneumoaiae, imipenem resistant Pseudomonas aerug- inosa and ESBL producing Escherichia coli. Methods: The water extracts of Coptidis Rhizoma were prepared firstly, and the disk agar plate diffusion method ( K-B method) was used to calculate the corresponding diameter of inhibition zone according to the related stand- ards in Clinical and Laboratory Standards Institute (CLSI). Besides, the broth micro-dilution method was used to figure out the mini- mum inhibitory concentration (MIC) of the water extracts of Coptidis Rhizoma. Results : According to the results of K-B method, Cop- tidis Rhizoma water extract had inhibitory effect on the four drug-resistant bacteria with the corresponding diameter of inhibition zone of 21 mm, 17 mm, 15 mm and 10 ram, respectively. The results of the broth micro-dilution method exhibited the bacteriostatic efficacy of Coptidis Rhizoma water extract on the four bacteria, and the MIC ofS. peneumoniae, MRSA, E. coli and P. aeruginosa was 15.63 mg · ml-1, 62.50 mg ·ml-1, 125 mg · ml-1 and 250 mg · ml-1, respectively. Conclusion: Coptidis Rhizoma water extract exhibits preferable baeteriostatic efficacy on the four common clinical infectious strains with the inhibitory effect order of ESBL producing S. pe- neumoniae 〉 MRSA 〉 ESBL producing E. coli 〉 imipenem resistant P. aeruginosa. The research shows significant value for the clinical treatment of bacterial infection diseases caused by S. peneumoniae and MRSA.
