摘要
目的:基于肝脏脂肪酸代谢探讨复方贞术调脂方(Fufang Zhenzhu Tiaozhi Formula,FTZ)对高脂饮食所致的脂代谢紊乱的作用及其可能机制。方法:C57BL/6J雄性小鼠随机分为正常对照组、模型组、二甲双胍阳性对照组、FTZ低、高剂量组;除对照组外,高脂饮食造模同时给药16周;检测小鼠空腹血糖(Fasted PlasmaGlucose,FPG)、甘油三酯(Triglyceride,TG)、总胆固醇(Total Cholesterol,TC)、低密度脂蛋白胆固醇(Low DensityLipoprotein-Cholesterol,LDL-C)、游离脂肪酸(Nonesterified Fatty Acid,NEFA)等血液指标,给药16周后进行口服糖耐量实验(Oral Glucose Tolerance Test,OGTT),并计算曲线下面积(Area Under The Curve,AUC);ELISA法检测血浆中空腹胰岛素(Fasted Insulin,FINS)浓度;HE染色、油红O染色观察肝脏病理情况;采用气相色谱-质谱联用(GC-MS)检测肝脏脂肪酸含量。结果:给药16周后,FTZ可以有效降低糖脂代谢紊乱小鼠血浆中FPG、TG、TC、LDL-C、NEFA和FINS浓度,并且可降低HOMA-IR值(Homeostasis Model Assessment-Insulin Resistent,HOMA-IR);OGTT结果显示FTZ可有效减少AUC值(P<0.05)。油红O染色结果显示FTZ可有效减少肝脏脂滴大小与数量。GC-MS结果显示FTZ可降低脂代谢紊乱小鼠肝脏总脂肪酸(Total Fatty Acids,TFA),其中主要的饱和脂肪酸(Saturated Fatty Acid,SFA)的含量下降,而大部分多不饱和脂肪酸(Polyunsaturated Fatty Acid,PUFA)的PUFA/TFA值有不同程度的升高。结论:FTZ可改善高脂饮食导致的糖脂代谢紊乱小鼠肝脏脂代谢,其机制与降低其肝脏中的SFA和升高肝脏中的PUFA相关。
This study was aimed to investigate the effect of Fufang Zhenzhu Tiaozhi Formula(FTZ) on lipid metabolism disorder mice induced by high fat diet and its possible mechanism based on fatty acid composition of liver. C57 BL/6 Jmale mice were randomly divided into 5 groups. The normal control group was given standard diet, and the rest group wasfed with high fat diet, while the administration group was prophylactic administration for 16 weeks. After 16 weeks oftreatment, oral glucose tolerance test(OGTT) was performed, and the area under the curve(AUC) was calculated, and thelevel of fasted plasma glucose(FPG), total cholesterol(TC), triglyceride(TG), low density lipoprotein-cholesterol(LDL-C) and nonesterified fatty acid(NEFA) were observed and recorded on a regular basis. The content of insulin in plasmawas detected by ELISA. The liver pathology was observed by HE staining and oil red O staining. The content of fattyacids in liver was determined by GC-MS. After 16 weeks of intragastric administration of FTZ, the results showed thatFTZ could effectively reduce the plasma concentrations of FPG, TG, TC, LDL-C, NEFA and insulin in mice with lipidmetabolic disorder induced by high fat diet. The OGTT showed that FTZ could effectively reduce the AUC value(P〈0.05). The results of oil red O staining showed that FTZ could effectively reduce the size and quantity of lipid droplets inliver. GC-MS results showed that FTZ could lead to significant decreases in saturated fatty acids(C14∶0, C15∶0, C16∶0 and C17∶0) in the mouse liver(P〈0.05). And FTZ can effectively increase the certain MUFA/TFA values such as C14∶1, C15∶1, C17∶1, C18∶1 N9 T, C20∶1, C24∶1, C22∶1 and certain PUFA/TFA values such as C20∶2, C22∶2, C18∶2 N6 T, C20∶3 N6, C20∶4 N6 and C22∶6 N3 to some extent(P〈0.05).
