麦冬寡糖对自发性2型糖尿病db/db小鼠降血糖的作用 被引量:13

摘要 为观察麦冬寡糖对自发性2型糖尿病db/db小鼠的降血糖作用,将40只db/db小鼠随机分为5组:模型对照组,阳性对照盐酸二甲双胍组,麦冬寡糖低、中、高剂量组,另设野生型db/m小鼠为正常对照组,分别灌胃给予相应药物或溶媒,连续8周。8周给药结束后,称量小鼠体质量,测定24 h尿蛋白量、24 h尿白蛋白量;给药4周、8周后,分别检测小鼠空腹血糖值(FBG),并于给药8周后进行口服糖耐量实验;最后,取小鼠血清,检测糖化血清蛋白(GSP)、三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)及血尿素氮(BUN)和血肌酐(Scr)含量。结果显示,麦冬寡糖能显著减轻自发性2型糖尿病db/db小鼠的体质量,降低空腹血糖值,减少口服葡萄糖耐量实验的曲线下面积并降低GSP含量;麦冬寡糖也能抑制24 h尿蛋白量、24 h尿白蛋白量、BUN和Scr含量的增加,显著降低血清TG、TC、HDL-C和LDL-C含量。综上所述,麦冬寡糖对自发性2型糖尿病db/db小鼠表现的肥胖、高血糖症、血脂异常、肾损伤均有较好的改善作用。 In order to investigate the hypoglycemic effect of Ophiopogonis japonicus oligosaccharide on type2diabetes mellitus,the db/db diabetic mice were intragastrically administered with Ophiopogonis japonicus oligosaccharide for8weeks.The body weight and general physical signs were observed during the treatment.The fasting blood glucose levels were determined after4and8weeks of administration.The changes in24h urinary albumin excretion and24h urine protein amounts,glycosylated serum protein(GSP),total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL C)and low density lipoprotein cholesterol(LDL C),urea nitrogen and creatinine levels in serum were measured after8weeks of treatment.Results showed that Ophiopogonis japonicus oligosaccharide could significantly lower the body weights,blood glucose levels and the AUCs during oral glucose tolerance test,and inhibited the changes of24h urinary albumin excretion and24h urine protein amounts,GSP,TC,TG,HDL C and LDL C,urea nitrogen and creatinine levels in serum of db/db diabetic mice by comparison with db/m mice.In conclusion,Ophiopogonis japonicus oligosaccharide exerts remarkable hypoglycemic and protective effects in db/db mice,thus showing its potential for use in the treatment of type2diabetes mellitus.
