摘要
目的建立白花蛇舌草的HPLC指纹图谱检测方法,对不同采收期的白花蛇舌草药材进行研究。方法色谱柱为Phenomenex C18柱(150mm×4.6mm,5μm),乙腈-2mL·L-1磷酸水溶液为流动相进行梯度洗脱,在203nm波长处以药材中的最高色谱峰为参照物测定了10批不同采收时间样品的指纹图谱。按照白花蛇舌草HPLC指纹图谱共有模式,标定16个共有指纹峰,并对不同采收批次样品指纹图谱进行相似度比较。结果同一产地的白花蛇舌草不同采收周期的药材有一定的差异,经比较从中选定了16个共有峰作为药材指纹图谱稳定的特征峰,共有峰占总峰面积的90%以上,不同采收期的药材与标准指纹图谱对照,相似度结果都在0.9以上。同时也对该指纹图谱检测方法进行了验证。结论通过HPLC指纹图谱法,能够控制白花蛇舌草药材的质量。
Objective To establish Hedyotis diffusa Willd. HPLC fingerprint and to identify the Hedyotis diffusa Willd. samples harvested at different times. Methods Phenomenex C18column (150 mm×4.6 mm,5μm) was used with acetonitrile-2 mL·L^-1phosphate solution as the mobile phase in gradient elution.10 different batch samples were analyzed under 203 nm and 16 common fingerprint peaks were employed to establish the model for the similarity comparison. Results The Hedyotis diffusa Willd. samples from same origin but different harvest times showed some differences.16 common peak fingerprints were selected as the characteristic peaks,accounted for more than 90% of the total peak area.The tested samples from different harvest times resulted in a similarity more than 0.9.At the same time,the fingerprint detection method was verified. Conclusion The HPLC fingerprint method can be used to control the quality of Hedyotis diffusa Willd..
