摘要
【目的】研究水性、油性及乳性基质对大鼠深Ⅱ度烫伤炎性期创面的影响。【方法】建立大鼠深Ⅱ度烫伤模型,创面外用地塞米松联合庆大霉素进行治疗,分别以磷酸盐缓冲液、大豆油和两者配成的乳剂作为基质,分为4组:空白组、水剂组、油剂组和乳剂组,每天给药2次,连续4 d。烫伤后不同时间点取材,紫外分光光度法测定超氧化物歧化酶(superoxide dismutase,SOD)含量,Realtime PCR检测肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)和白细胞介素6(interleukin-6,IL-6),ELISA检测IL-1蛋白表达,伤口石蜡切片行HE染色观察炎症反应和组织修复情况。【结果】在炎性期,水、油、乳基质抗炎效果与空白组相比较差,各溶剂组SOD含量均低于空白组。HE染色发现烫伤后细胞间水肿和呈空泡化,在血管周围有炎细胞浸润,油剂组最为明显(第4天)。水剂能够提高炎症因子TNF-α、IL-6的基因表达和IL-1蛋白表达。【结论】不同溶剂不利于烧伤后的抗氧化应激作用,其中水剂基质可引发较重炎症反应。
【Objective】To study the effects of water, oil and emulsion matrix on the deep second degree scalded wounds of rats in the inflammatory phase.【Methods】The rat model with deep second degree scalded wounds was established. The external wounds were treated with dexamethasone combined with gentamicin, which was mixed with emulsion matrix prepared from phosphate buffer solution, soybean oil or both, 2 times a day for 4 days. The materials were taken at different time points after the scald. The content of superoxide dismutase(SOD) was measured by UV spectrophotometric method. The expressions of tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6) were detected by Realtime PCR. The expression of interleukin-1(IL-1) was determined by ELISA. The wound Inflammation reaction and tissue repairation were observed by HE staining.【Results】In the inflammatory stage, the anti-inflammatory effects of water, oil and emulsion matrix were worse than that in the blank group, and the SOD content in each solvent group was lower than that in the blank group. The results of HE staining showed that the intercellular edema and vacuolization were found in scalded rats, and inflammatory cells were infused around the blood vessels, which was the most obvious in the oil group on the fourth day. Water matrix could increase the gene expressions of TNF-α, IL-6 and protein expression of IL-1.【Conclusion】Different solvents are not conducive to the anti-oxidative stress after scald, in which water matrix can cause heavy inflammatory responses.
