摘要 目的研究Survivin基因及蛋白表达与人786-0肾透明癌细胞增殖和凋亡的相关性。方法人肾癌细胞系786-0作为Survivin转染组,肾小管上皮细胞系HK-2作为阴性对照组,分别用Survivin模拟物转染;转染72 h后测定Survivin基因及蛋白表达;对比两组细胞吸光值、细胞数及凋亡细胞比值;对Survivin基因和蛋白表达水平与细胞吸光值、细胞数和凋亡细胞比值进行Pearson相关性分析。结果转染后,Survivin转染组Survivin mRNA、Survivin蛋白表达水平显著高于阴性对照组(P<0.05);吸光值从第1 d开始显著低于阴性对照组(P<0.05);细胞数从第2d开始显著少于阴性对照组(P<0.05)。786-0细胞的凋亡比例从第1d开始显著低于阴性对照组(P<0.05)。Pearson相关性分析结果显示,Survivin基因和蛋白表达水平与吸光值和细胞数呈负相关,与凋亡细胞比值呈正相关。结论 Survivin基因和蛋白表达可以抑制786-0肾透明癌细胞增殖,促进细胞凋亡。 Objective To study the correlation between Survivin gene and protein expression and proliferation and apoptosis of human 786-0 renal clear cancer cells. Methods Human renal cell carcinoma cell line 786-0 was used for survivin transfection group, while renal tubular epithelial cell HK-2 was used as negative control group and transfected with survivin mimic. The expression levels of survivin gene and protein was determined 72 h after transfection. Absorbance value, cell number and ratio of apoptotic cells in two groups were compared; Pearson correlation analysis was conducted based on Survivin gene and protein expression levels and cell light absorbance value, cell number and apoptotic cell ratio. Results The expressions of Survivin mRNA and Survivin protein in Survivin transfection group were significantly higher than that in negative control group ( P 〈 0.05 ). The absorbance value of Survivin transfection group was significantly lower than that in negative control group ( P 〈 0.05 ). From the 2nd day, the number of cells in transfection group was significantly lower than the negative control group( P 〈 0.05 ). The percentage of apoptosis in 786-0 cells was significantly lower than that in the negative control group from the first day (P 〈 O. 05 ). Pearson correlation analysis showed that the expression levels of Survivin gene and protein were inversely proportional to the absorbance value and cell number, and were positively proportional to the ratio of apoptotic cells. Conclusion Survivin gene and protein expressions can inhibit the proliferation of 786-0 renal clear cancer cells and promote apoptosis.
