Ad-pre-microRNA-494抑制Survivin对膀胱癌UM-UC-3细胞的凋亡诱导作用 被引量:1

摘要 目的观察腺病毒载体Ad-pre-microRNA-494抑制Survivin表达对人膀胱尿路上皮癌UM-UC-3细胞周期及凋亡的影响。方法使用已构建的腺病毒载体Ad-pre-microRNA-494感染人膀胱尿路上皮癌UM-UC-3细胞,采用RT-PCR法、Western blot法、流式细胞计数法检测感染前后Survivin基因及蛋白的表达差异和细胞生长周期及凋亡情况。结果实验组细胞与空病毒载体组细胞相比较,Survivin基因表达下调,细胞凋亡率显著增加(P<0.01)。结论 Ad-premicroRNA-494能有效抑制UM-UC-3细胞中Survivin基因表达,从而诱导UM-UC-3细胞凋亡,这为下一步的动物实验研究奠定了基础。 Objective To investigate the effect of adenovirus vector Ad-pre-microRNA-494 on the inhibition of Survivin gene expression and the apoptosis induction of UM-UC-3 cells. Methods Adenovirus vector Ad-pre-microRNA-494 was constructed to infect human bladder transitional cell carcinoma UM-UC-3 cells.RT-PCR,Western blot and flow cytometry assay were used to detect the expression of Survivin gene,cell growth cycle and apoptosis and compare the difference between before and after infection. Results The expression of Survivin gene in the experimental group downregulated compared with empty vector group,and the apoptosis rate was significantly increased(P<0.01). Conclusions Ad-pre-microRNA-494 can effectively inhibit the expression of Survivin gene in UM-UC-3 cells and induce the apoptosis of UM-UC-3 cells,which lays the foundation for the further animal experiment.
