MUC2和TFF3在结肠腺瘤和腺癌组织的表达及意义 被引量:3

Expressions and clinical significance of MUC2 and TFF3 in human colorectal adenoma and adenocarcinoma

摘要 目的检测肠杯状细胞标志物黏蛋白2(MUC2)和三叶因子3(TFF3)在人正常结肠、结肠腺瘤和腺癌组织的表达,并探讨其临床意义。方法收集正常结肠组织标本10例、结肠腺瘤15例以及结肠腺癌50例,采用免疫组化染色SP法检测MUC2和TFF3的表达,并分析其与临床病理特征的关系。结果正常结肠、结肠腺瘤及腺癌组织的MUC2表达阳性率分别为:100%(10/10)、73.3%(11/15)、38.0%(19/50);TFF3表达阳性率分别为:100%(10/10)、66.6%(10/15)、44.0%(22/50),二者表达均逐渐降低,不同组织间比较均差异显著(P<0.05)。MUC2和TFF3的表达均与结肠癌患者的年龄、性别、癌发生部位及分化程度无关(P>0.05),而与结肠癌的TNM分期、淋巴结及远处转移相关(P<0.05)。结论在结肠"正常黏膜-腺瘤-腺癌"的演变过程中,MUC2和TFF3表达逐渐减少,向正常的肠杯状细胞分化减少,二者的联合检测可能有助于对结肠癌变的早期预测;且该表达减少与结肠癌的TNM分期、淋巴结及远处转移相关,二者可能作为判断结肠癌进展和转移的预测因子。 Objective To detect the expressions of intestinal goblet cell markers mucin 2( MUC2) and trefoil factor 3( TFF3) in normal colon,colon adenoma and adenocarcinoma tissues,and further explore their clinical significance. Methods Ten cases of normal colon tissue,15 cases of colon adenoma and 50 cases of colon adenocarcinoma were collected. The expressions of MUC2 and TFF3 were detected by immunohistochemical staining( SP),and the relationship between their expressions and clinicopathological characteristics were analyzed.Results In the normal colon,colonic adenoma and adenocarcinoma,the positive rate of MUC2 expressions was100%( 10/10),73. 3%( 11/15) and 38%( 19/50),respectively; and the positive rate of TFF3 expressions was 100%( 10/10),66. 6%( 10/15) and 44%( 22/50),respectively. The expressions of MUC2 and TFF3 were gradually decreased. There was significantly difference between tissues( P〈0. 05). The expressions of MUC2 and TFF3 were not related to the age,sex,location and tumor differentiation degree of colon cancer patients( P〉0. 05),but were correlated with TNM stage,lymph node and distant metastasis of colon cancer( P〈0. 05). Conclusion In the evolution of " normal colon mucosa-adenoma-adenocarcinoma",the expressions of intestinal goblet cell markers MUC2 and TFF3 are gradually decreased,which suggests a decrease in the differentiation of normal intestinal goblet cells is shown. The combined detection of MUC2 and TFF3 might contribute to the early prediction of colon carcinogenesis. The decreased expressions are related to the TNM stage,lymph node and distant metastasis of colorectal cancer,which implies that MUC2 and TFF3 might be the predictors of progression and metastasis of colorectal cancer.
