杏扁品种‘优一’组织培养再生体系建立的初步研究 被引量:3

摘要 目的建立杏扁品种‘优一’的组培快繁技术体系。方法选择‘优一’生长健壮、一年生枝的带芽茎段作为外植体,对其初代诱导的外植体采集时间、灭菌时间、基本培养基的选择以及不同激素浓度对继代培养和生根培养的影响进行优化研究。结果外植体的最佳采集时间应在5月;表面消毒利用0.1%HgCl2溶液灭菌8min效果较好,污染率为21.7%,萌芽率为65.0%;茎段起始培养选用MS为基本培养基,附加1.0mg·L^(-1) 6-BA和0.1mg·L^(-1) NAA,增殖芽数较多,萌芽率高。适宜的增值培养基为MS+6-BA1.0mg·L^(-1)+NAA0.1mg·L^(-1),平均增值芽数为4.1,芽苗生长较壮,茎芽长度为3.30cm;生根培养基可选择1/2MS+IBA0.4mg·L^(-1),平均生根数为2.4,生根率为70.0%。结论建立了杏扁品种‘优一’的组培快繁体系,为培育健康优良种苗提供了实用指南,有利于杏扁产业的可持续发展。 Objective To establish tissue culture and rapid propagation system of Prunus armeniac.Methods By using ‘Youyi'stems with buds and strong annual branches,as explants,the primary explant induction on the acquisition time,sterilization time,basic medium and different hormone concentration on the effects of subculture and rooting culture were studied.Results May was the optimum time to pick explant.The most appropriate sterilization method of explant was sterilization for 8 min in 0.1%HgCl2 solution,the pollution rate was 21.7%,and the germination rate was 65%.Adding 1.0 mg/L 6-BA and 0.1 mg/L NAA in MS medium was better than others for the beginning culture,and it improved buds induce and growth.The suitable medium of subculture proliferation was MS+6-BA1.0 mg/L+NAA0.1 mg/L,the average increment bud number was 4.1,the bud growth was stronger,and the stem bud length was 3.30 cm;the rooting medium was 1/2 MS+IBA0.4 mg/L,the average number of roots was 2.4,and the rooting rate was 70%.Conclusion Tissue culture and rapid propagation system of Prunus armeniac was established,which provided a practical guide for the cultivation of healthy seedlings,and conducived to the sustainable development of the almond industry.
