摘要
目的同时测定大鼠血浆中木樨草素和牡荆素的含量,考察同时测定木犀草素和牡荆素的高效液相色谱方法。方法采用高效液相色谱法,色谱柱Diamonsil ODS C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-20 mmol/L磷酸二氢钾缓冲液(58:42,pH=4.0),检测波长350 nm,流速为1.0 m L/min,柱温30℃。结果分别在38.5~4 350 ng/m L和16.5~1 860 ng/m L范围内,木樨草素和牡荆素的校准曲线呈线性相关,相关性系数r为0.999 6和0.999 4,方法回收率分别为102.3%~104.7%和93.82%~101.4%,日内和日间精密度(RSD)小于7.9%和5.4%。结论建立的高效相色谱方法稳定,结果可靠,可用于血浆中木樨草素和牡荆素浓度的检测。
Objective To determine the contents of luteolin and vitexin in rat plasma,and measure the HPLC method of luteolin and vitexin simultaneously. Methods HPLC method was used. Diamonsil ODS C18 column was( 250 mm × 4. 6 mm,5 μm),the mobile phase was methanol-20 mmol/L potassium dihydrogen phosphate buffer solution( 58 ∶ 42,pH4. 0),the detection wavelength was 350 nm,the flow rate was 1. 0 m L ·min,and the column temperature was 30 ℃. Results The standard curve for luteolin and Vitexin in plasma were linear over the range of 38. 5 ~ 4 350 ng/m L and 16. 5 ~ 1 860 ng/m L. The calibration curves of luteolin and Vitex showed linear correlation,with the correlation coefficients being 0. 999 6 and 0. 999 9 respectively. The assay recoveries for luteolin and Vitexin ranged from 102. 3% to 104. 7% and from 93. 82% to 101. 4% respectively. The intra-and inter-day precisions( RSD) for luteolin and vitexin were all less than 7. 9% and 5. 4%. Conclusion The HPLC method is stable and reliable,which can be used for the determination of luteolin and vitexin in plasma.
