摘要
为验证丝素蛋白轻链(Fib-L)基因对棉大卷叶螟[Sylepta derogata(Fabricius)]丝素蛋白的影响,采用RNA干扰(RNAi)技术沉默Fib-L基因,系统测定棉大卷叶螟5龄幼虫丝中氨基酸组分和丝的力学性能。结果表明:棉大卷叶螟Fib-L基因被干扰后,氨基酸组分与对照相比发生了明显变化,特别是酪氨酸经RNAi处理后几乎检测不到;碱性与酸性氨基酸及小侧基与大侧基氨基酸比例也明显下降;RNAi处理后棉大卷叶螟丝的吸湿能力受到明显影响,且较大幅度地降低丝的力学性能。该研究提示Fib-L基因在调控棉大卷叶螟吐丝过程中起着重要作用。
Sylepta derogata (Fabricius) is the primary pest and causes grave damage on cotton as well as other crops in the middle and lower reaches of the Yangtze river by direct feeding and indirect curling and spinning at larval stage. In this study, to clarify the influence of fibroin light chain (Fib-L) gene on the fiboin protein, the amino acid composition and silk mechanical property of silk produced by the 5th instar larvae were determined after Fib-L gene was silenced by RNA interference (RNAi) technique. The results showed that the composition of the amino acids was significantly different compared to the control. The tyrosine was barely detected. In addition, the proportion of alkaline amino acids to acidic amino acids as well as amino acids with small-side group to bulk-side group decreased significantly in RNAi treatment. Meanwhile, the moisture absorption capacity of silk was affected and the elasticity of silk was also reduced. The results indicate that the Fib-L gene plays a key role in silk-spinning of S. derogata.
