摘要
目的观察不同浓度梯度的舒芬太尼体外处理人肺腺癌细胞A549后对细胞增殖和凋亡的影响。方法采用CCK-8法检测舒芬太尼(0.01、0.02、0.04μg/m L)处理A549细胞48 h后的增殖情况,采用Annexin V-FITC/PI双染法检测舒芬太尼处理A549细胞48 h后的细胞凋亡情况,Western blotting检测各浓度舒芬太尼作用A549细胞48h后的凋亡相关蛋白p53、Bcl-2和Bax的表达水平。结果舒芬太尼能够抑制A549细胞增殖以及诱导其凋亡,且增值率和凋亡率的变化呈剂量依赖性(P<0.05)。与对照组相比,各浓度舒芬太尼组A549细胞的凋亡率均有不同程度升高(P<0.05),且p53和Bax蛋白水平表达升高,而Bcl-2蛋白表达量降低(P<0.05)。结论舒芬太尼可抑制人肺腺癌A549细胞增殖并促进其凋亡,可能与舒芬太尼能够上调p53和Bax蛋白的表达以及下调Bcl-2蛋白的表达有关。
Objective To explore the effects of sufentanil on the proliferation and apoptosis of A549 cells. Methods The CCK-8 was used to detect the proliferation of A549 cells at 48 h after treatment with different concentrations of sufentanil( 0. 01,0. 02,0.04μg/m L). Annexin V-FITC/PI double staining was used to detect the apoptosis rate after treatment with different concentrations of sufentanil. Western blotting was used to detect the expression levels of apoptosis-related protein p53,Bcl-2 and Bax after treatment with different concentrations of sufentanil. Results Sufentanil could inhibit the proliferation and promote the apoptosis of A549 cells at different concentrations in dose-dependently manner( P 〈 0. 05). Compared with the control group,the apoptotic rate of A549 cells was increased after treatment with different concentrations of sufentanil( P 〈 0. 05). The treatment with sufentanil leaded to the increased levels of p53 and Bax but decreased level of Bcl-2( P 〈 0. 05). Conclusion Sufentanil could inhibit proliferation and promote the apoptosis of A549 cells,possibly via the upregulation of p53 and Bax and downregulation of Bcl-2.
