摘要
目的探讨丁香酚吸嗅对大鼠脑缺血/再灌注(MCAO)损伤后空间学习记忆能力及海马CA_3区HE染色神经元影响。方法线栓法建立大鼠MCAO模型。将40只SD雄性大鼠用数学表法随机分为正常组、假手术组(SH组)、模型组(MCAO)(I/R组)、丁香酚吸嗅组(丁香酚吸嗅+MCAO)(Egu组)。经丁香酚吸嗅干预,2 W后通过Morris水迷宫检测再灌注后大鼠的空间学习记忆能力;HE染色检测海马CA3区HE染色神经元变化。结果与模型组相比,丁香酚吸嗅组学习能力和记忆能力较模型组得到明显改善(P<0.01);HE染色显示,丁香酚吸嗅组大鼠海马CA_3区神经细胞较模型组数量增多,排列较整齐;且细胞形态、结构较完整。结论丁香酚吸嗅可以改善脑缺血/再灌注大鼠空间学习记忆能力,HE染色显示对大鼠海马CA_3区神经元具有保护作用。
Objective To investigate eugenol sniff ing on cerebral ischemia/reperfusion injury in spatial learning and memory ability and effect of HE staining on hippocampal CA3 neurons.Methods The rat MCAO model was established by suture method. The 40 male SD rats were randomly divided into normal group、sham group(SH group) 、 model group(MCAO)(I/R group)、 eugenol sniffing group(eugenol sniffing + MCAO)(Egu group).By the intervention of eugenol sniffing, 2 W late we detected rat spatial learning and memory ability by Morris water maze after reperfusion;Changes of HE staining in hippocampal CA3 region by HE staining. Results Compared with the model group, eugenol sniffing group learning ability and memory improved significantly comparing with the model group(P 0.01); HE staining showed that eugenol sniffing rats increased number of cells in the hippocampal CA3 region comparing with the model group, arranged in neat; and cell morphology, structure is more complete. Conclusion Eugenol sniffing can improve cerebral ischemia/reperfusion in spatial learning and memory in rats and HE staining showed protective effect on hippocampal CA3 neurons in rats.
