摘要
目的:探讨丁香酚吸嗅对大鼠脑缺血/再灌损伤后空间学习记忆能力及海马区5-羟色胺(5-HT)表达的影响。方法:线栓法建立大鼠脑缺血/再灌模型。采用数字表法将40只SD雄性大鼠随机分为假手术组、正常组、缺血/再灌注组、丁香酚吸嗅组。经丁香酚吸嗅干预,各组大鼠于2周后通过Morris水迷宫测试定位航行试验和空间探索试验,检测再灌注后大鼠的空间学习记忆能力;免疫组化观察大鼠海马区5-HT表达。结果:与缺血/再灌注组相比,丁香酚吸嗅组学习能力和记忆能力较缺血/再灌注组得到明显改善(P<0.01);免疫组化实验结果示,丁香酚吸嗅组大鼠海马区5-HT神经递质较缺血/再灌注组有明显增加(P<0.01)。结论:丁香酚吸嗅可以改善脑缺血/再灌大鼠空间学习记忆能力,其作用机理可能与大鼠海马区5-HT的改变有关。
Objective:To investigate eugenol sniffing on cerebral ischemia/reperfusion injury in spatial learning and memory ability and hippocampal 5-HT expression.Methods: The rat model of cerebral ischemia/reperfusion was established by suture method. The 40 male SD rats were randomly divided into normal group, sham group, ischemia/reperfusion group, eugenol sniffing group.The eugenol sniffing intervention after 2 w by Morris water maze test positioning navigation test and spatial probe test.Detection after reperfusion rat spatial learning and memory ability;Immunohistochemical observation rat hippocampus 5-HT expression.Results: Compared with the ischemia/reperfusion group, eugenol sniffing group learning ability and memory improved significantly comparing with the ischemia/reperfusion group(P〈0.01);The results of immunohistochemistry showed that 5-HT neurotransmitter in hippocampus of eugenol group was significantly higher than that in ischemia/reperfusion group(P〈0.01). Conclusion: eugenol sniffing can improve cerebral ischemia/reperfusion in spatial learning and memory in rats and its mechanism may be related to changes in 5-HT in the hippocampus of rats related.
