摘要
为了研究10种中药发酵制剂对过氧化氢(H2O2)诱导石斑鱼肝细胞氧化损伤的保护作用,试验用过氧化氢和10味中药发酵制剂同时处理斜带石斑鱼肝细胞8 h后测定细胞存活率及培养上清液中的天门冬氨酸氨基转移酶(AST)、谷氨酸氨基转移酶(ALT)、乳酸脱氢酶(LDH)活性;同时测定细胞中的总抗氧化能力(T-AOC)、丙二醛(MDA)含量以及谷胱甘肽过氧化物酶(GSH-Px)活性。结果表明:在H2O2的作用下,细胞存活率显著下降(P〈0.05),AST、ALT和LDH活性显著升高(P〈0.05),T-AOC和GSH-Px活性显著降低(P〈0.05);在10种中药发酵制剂中,银杏叶和甘草两种发酵制剂的AST和ALT活性都显著低于其他各组(P〈0.05),T-AOC、GSH-Px活性显著高于其他各组(P〈0.05),且MDA含量在各组中最低(P〈0.05)。说明与其他药发酵制剂相比,银杏叶与甘草发酵制剂对H2O2诱导的石斑鱼肝细胞氧化损伤有着较强的保护作用。
In order to research protective effect of 10 kinds of fermented traditional Chinese medicine( TCM) against oxidative damage of grouper liver cells induced by H2 O2,H2 O2 and 10 kinds of fermented TCM were used in this experiment simultaneously in the treatment of Saddletail grouper liver cells. Eight hours post treatment,cell viability was determined,also including the activities of aspartate aminotransferase( AST),alanine transaminase( ALT),lactic dehydrogenase( LDH); meanwhile total antioxidant capacity( T-AOC),the content of Malondialdehyde( MDA),and the activity of glutathione peroxidase( GSH-Px) in liver cells were also measured. The results showed that under the action of H2 O2,cell viability reduced significantly( P〈0. 05),while the contents of AST,ALT and LDH increased significantly( P〈0. 05) and T-AOC and GSH-Px activities decreased significantly( P〈0. 05). Among 10 kinds of fermented TCM,AST and ALT contents in Ginkgo biloba group or Liquorice group were significantly lower than those in other groups( P 0. 05),while T-AOC and GSH-Px activities were significantly higher than those in other groups( P〈0. 05),and the MDA content was the lowest in each group( P〈0. 05). The results suggested that compared with other fermented TCM,Ginkgo biloba and Liquorice could have a strong protective effect against oxidative damage of grouper liver cells induced by H2 O2.
