摘要
目的探讨厚叶南五味子醇提物(KCAE)抗大鼠免疫性肝纤维化作用,并对其机制做初步研究。方法 SD雄性大鼠60只随机分为正常对照组、模型组、复方鳖甲软肝片组(0.7 g·kg^(-1))、KCAE高、中、低剂量组[1.68、0.84、0.42 g(药材)·kg-1],每组10只。除正常对照组外,其余各组均腹腔注射猪血清诱导大鼠肝纤维化模型,0.5 m L·只^(-1),每周2次。造模同时各给药组分别给予相应药物,正常对照组和模型组予等量生理盐水,每天1次,共15周。测定大鼠肝脏指数,生化检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总蛋白(TP)、白蛋白(ALB)、总胆红素(TB)含量变化,酶联免疫吸附法(ELISA)测定血清中Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(Ⅳ-C)、层粘连蛋白(LN)、透明质酸酶(HA)、生长转化因子-β_1(TGF-β_1)、白介素-10(IL-10)、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)含量,苏木素-伊红(HE)染色、马松(Masson)染色观察大鼠肝纤维化程度,Western blot法检测肝组织中TGF-β_1蛋白的表达。结果与模型组比较,KCAE高剂量可显著降低大鼠肝脏指数(P<0.01);各组大鼠血清ALT、AST、TP、ALB、TB水平均位于正常范围,差异无统计学意义(P>0.05);KCAE可以降低血清中PCⅢ、IV-C、LN、HA、TGF-β_1及TNF-α含量,提高IFN-γ水平;KCAE可显著改善大鼠肝纤维化程度(P<0.01),明显抑制肝组织TGF-β1的表达(P<0.01)。结论 KCAE具有显著的抗大鼠免疫性肝纤维化作用,其机制可能与其调控炎症细胞网络、减少细胞外基质及抑制TGF-β1表达有关。
OBJECTIVE To study the effects of Kadsura coccinea alcohol extract(KCAE) on rats with immunologic hepatic fibro- sis and research the possible mechanisms in it. METHODS Totally 60 SD male rats were randomly divided into 6 groups : a normal control group, a model group, a compound Biejia-ruangan tablets group ( 0. 7 g · kg^-1 ) , KCAE high, middle and low dose groups ( 1.68, 0. 84, 0. 42 g · kg^-1 ) at ten in each groups. Except for the normal control group, other groups were duplicated intraperitoneal injection of porcine serum twice a week at dose of 0. 5 mL · time^-1. The rats in treatment groups were intragastric administration re- spectively, meanwhile, the rats in normal control and model groups were treated with the same volume of distilled water, once a day for 15 weeks. The liver was weighed to calculate the liver index. Alanine aminotransferase( ALT), aspartate aminotransferase( AST), total protein( TP), albumin(ALB) and total bilirubin(TB) were evaluated by the Mind-Ray automatic biochaemical analyzer. The expres- sion level of procollagen m ( PC m ), collagen type IV ( IV-C ), laminin ( LN ), hyaluronic acid ( HA ) , transforming growth factor-131 (TGF-β1), interkeukin-10(IL-10), interferon-γ (IFN-γ) and tumor necrosis factor-α(TNF-α) in serum were detected by ELISA. The degrees of fibrosis were evaluated by HE and Masson straining, and the expression levels of TGF-γ in liver tissue were assessed by Western blot. RESULTS Compared with model group, the liver index of KCAE high-dose group was decreased significantly ( P 〈 0. 01 ). The expression levels of ALT, AST, TP, ALB, TB were within normal range, the differences were not statistically significant (P 〉0. 05). KCAE could decrease the level of Pcm, iv-c, LN, HA, TGF-β 1, TNF-α and increase the level of IFN-γ in serum. KCAE could alleviate the hepatic fibrosis in rats( P 〈 0. 01 ) and inhibit the expression of TGF-β1 in the liver tissues significantly( P
