摘要
目的:探讨细胞块技术检测宫颈液基脱落细胞(LCT)中抑癌基因P16、抑癌基因IMP3和Stathmin蛋白表达在宫颈病变诊断的意义和价值。方法:采用免疫组化法检测146例宫颈脱落细胞块中P16、IMP3以及Stathmin的表达情况。结果:随着病变级别的升高而P16、IMP3和Stathmin阳性表达率和强表达率有升高的趋势,其在高级别鳞状上皮内病变(HSIL)组、鳞状上皮细胞癌(SCC)组的阳性表达率显著高于其他组(P<0.05);SCC组、HSIL组、低级别鳞状上皮内病变(LSIL)组与未见上皮内病变(NILM)组比较,P16、IMP3和Stathmin蛋白3指标的同一指标阳性率比较差异均有统计学意义(P<0.05)。在HSIL组和SCC组病例中,P16、IMP3和Stathmin三者表达呈正相关。结论:将LCT标本行细胞块制作、联合检测P16、IMP3和Stathmin表达,可能能提高宫颈癌筛查的准确性,为指导治疗及评估预后的进一步研究奠定了基础。
Objective:To detect the expression of P16,IMP3 and Stathmin in cervical exfoliated cells by applying the cell block preparations and to investigate the pathological significance in the use of diagnoses in cervix disease. Methods: Immunohistochemical method was used to detect the expression of p16, IMP3 and Stathmin in cervical deciduous cells from 146 cases. Results:The positive expression rate and strong expression rate of p16, IMP3 and Stathmin were correlated with the increasing grades of the diseases. The positive expression rate in HISL and SCC group was higher than that in other groups(P〈0.05). Compared with NILM group,the positive expression rates of the three proteins in SCC, HISL and LISL group had statistical significances ( P 〈 0.05). In HISL and SCC groups,there was a positive correlation among p16,1MP3 and Stathmin protein expression. Conclusions:Combining the making of cervical celt block with the detection of p16,tMP3 and Stathmin protein could improve the accurate detection rate of cervical cancer and direct treatment and prognostic evaluation.
