柴胡三参胶囊对p38MAPK通路介导BMSCs向心肌细胞分化的干预研究 被引量:8

摘要 目的研究柴胡三参胶囊含药血清干预骨髓间充质干细胞在p38MAPK通路向心肌细胞分化的作用,探讨其抗心律失常机制与p38MAPK信号转导通路的相关性。方法根据不同处理因素将骨髓间充质干细胞分为4组:空白对照组、BMP-2诱导剂组、柴胡三参含药血清+BMP-2诱导剂组、柴胡三参含药血清组,分别加入空白血清、BMP-2、柴胡三参含药血清+BMP-2及柴胡三参含药血清,用WB法检测各组内p38MAPK m RNA表达水平,RT-PCR法检测干预后各组中MEF2C、CTn T蛋白表达。结果 WB法结果显示:与其他各组对照,柴胡三参+BMP-2组p38MAPK m RNA表达均上调(P<0.01);以最佳浓度即20%的含药血清给予柴胡三参+BMP-2组、柴胡三参组干预后,空白组、BMP-2组、柴胡三参组p38MAPK m RNA表达较柴胡三参+BMP-2组均见下调(P<0.05或P<0.01),而空白组表达与其他组比较均无统计学意义。RT-PCR结果显示:诱导后BMP-2组、BMP-2+柴胡三参组、柴胡三参组MEF2C及CTn T表达水平均高于空白血清组,差异有统计学意义(P<0.01);BMP-2+柴胡三参组表达水平高于BMP-2组、柴胡三参组(P<0.05);BMP-2组与柴胡三参组比较,差异无统计学意义(P>0.05)。结论柴胡三参胶囊含药血清在一定条件下能促进骨髓间充质干细胞的增殖,对P38MAPK通路介导骨髓间充质干细胞向心肌细胞分化具有一定的促进作用。 Objective: To study the effects of Chaihu Sanshen Capsule(CHSSC) medicated serum on the differentiation of BMSCs into Myocardial Cell by P38 MAPK signal pathway and to explore the relationship between the antiarrhythmic mechanism and P38 MAPK signal transduction pathway. Methods: Bone marrow mesenchymal stem cells were divided into 4 groups according to different treatment factors: the blank control group,BMP-2 group,CHSSC+BMP-2 group and CHSSC group. The blank serum,BMP-2,CHSSC+BMP-2 and Chaihu Sanshen Capsule(CHSSC) medicated serum were given to corresponding groups respectively. The m RNA expression of p38 MAPK was detected with WB in each group;the protein expression of MEF2 C and CTn T after intervention was detected with RT-PCR in each group. Results: WB results showed that compared with other groups,the m RNA expression of p38 MAPK in CHSSC+BMP-2 group increased(P〈0.01). After intervention by the best concentration containing plasma(20%),the m RNA expression of p38 MAPK in the blank control group,BMP-2 group and CHSSC group decreased(P〈0.05 or P〈0.01),but the blank group showed no significant difference compared with other groups. RT-PCR results showed that after induction,the expression levels of MEF2 C and CTn T in BMP-2 group,CHSSC +BMP-2 group and CHSSC group were higher than those in blank serum group,and the difference was statistically significant(P〈0.01);the levels of CHSSC+BMP-2 group was higher than that of BMP-2 group and CHSSC group(P〈0.05). There was no statistical significance between BMP-2 group and CHSSC group(P〉0.01). Conclusion: CHSSC medicated serum can promote the proliferation of bone marrow mesenchymal stem cells under certain conditions and promote the differentiation of mediated bone marrow mesenchymal stem cells to cardiomyocytes by p38 MAPK pathway.
