柴胡三参胶囊对缺血性心律失常大鼠心肌组织CaMKⅡ蛋白及CaMKⅡ-mRNA表达的影响 被引量:2

摘要 目的探讨柴胡三参胶囊抗缺血性心律失常的作用及分子学作用机制。方法将60只SPF级雄性Wistar大鼠随机分为柴胡三参胶囊组、参松养心胶囊组、维拉帕米组、假手术组、模型组和空白组,每组10只。柴胡三参胶囊组、参松养心胶囊组、维拉帕米组预防性给予相应药物灌胃,假手术组、模型组和空白组灌胃等量的生理盐水,连续灌胃2周后,柴胡三参胶囊组、参松养心胶囊组、维拉帕米组大鼠均复制缺血性心律失常模型,假手术组大鼠只穿线不结扎,观察各组大鼠ECG变化,Western blot法检测心肌组织中CaMKⅡ蛋白表达水平,定量PCR法检测心肌组织中CaMKⅡmRNA表达水平。结果模型组大鼠房性早搏、室早、室速、室颤发生率及心律失常积分、心肌组织中CaMKⅡ蛋白及CaMKⅡmRNA表达水平均显著高于空白组(P均<0.05);柴胡三参胶囊组、参松养心胶囊组、维拉帕米组室速、室颤发生率及心律失常积分、心肌组织中CaMKⅡ蛋白及CaMKⅡmRNA表达水平均显著低于模型组(P均<0.05);柴胡三参胶囊组及参松养心胶囊组室速、室颤发生率心律失常积分、心肌组织CaMKⅡ蛋白及CaMKⅡmRNA表达水平均显著低于维拉帕米组(P均<0.05)。结论柴胡三参胶囊能有效降低室速、室颤发生率,并可减轻心肌缺血性心律失常造成的心肌组织损伤,其可能通过抑制CaMKⅡ蛋白和CaMKⅡmRNA的表达而发挥心肌保护作用。 Objective It is to explore the effect and molecular mechanism of Chaihu Sanshen capsules against ischemic arrhythmia.Methods Sixty SPF male Wistar rats were randomly divided into Chaihu Sanshen capsule group,Shensong Yangxin capsule group,verapamil group,sham operation group,model group and blank group,10 rats in each group.Chaihu Sanshen capsule hroup,Shensong Yangxin capsule group and verapamil group were given the preventive corresponding medicine by gavage,and sham operation group,model group and blank group were given the same amount of normal saline.After 2 weeks of gavage,the ischemic arrhythmia models were replicated in the Chaihu Sanshen capsule group,Shensong Yangxin capsule group and verapamil group.The rats in the sham operation group were only threaded without ligation.The changes of ECG of the rats in every group were observed,the expression level of CaMKⅡprotein in myocardial tissue was detected by blot method,and the expression level of CaMKⅡmRNA in myocardial tissue was detected by quantitative PCR method.Results The occurrence rates of atrial premature beats,ventricular premature beats,ventricular tachycardia,ventricular fibrillation and arrhythmia scores,the expression levels of CaMKⅡprotein and CaMKⅡmRNA in myocardial tissue were significantly higher in the model group than those in the blank group(all P<0.05);the occurrence rates of ventricular tachycardia,ventricular fibrillation and arrhythmia score,expression of CaMKⅡprotein and CaMKⅡmRNA in myocardial tissues of Chaihu Sanshen capsule group,Shensong Yangxin capsule group and verapamil group were significantly lower than those of model group(all P<0.05);the occurrence rates of ventricular tachycardia,ventricular fibrillation and arrhythmia score,expression of CaMKⅡprotein and CaMKⅡmRNA in myocardial tissues of Chaihu Sanshen capsule group and Shensongyangxin capsule group were significantly lower than those in verapamil group(all P<0.05).Conclusion Chaihu Sanshen capsule can effectively reduce the incidence of ventricular t
