氧化苦参碱对大鼠缺血再灌注心肌损伤生化指标的影响 被引量:23

摘要 目的:探讨氧化苦参碱对大鼠缺血再灌注心肌的保护作用。方法:45只成年大鼠随机分成3组:正常对照组、缺血再灌注组、氧化苦参碱组,麻醉大鼠后冠脉结扎30min,再灌60min后,各组立即取出心脏,用冰盐水冲洗、吸干、称重后以生理盐水为介质,用匀浆器制成心肌组织匀浆,观察3组大鼠心肌组织匀浆中脂质过氧化物质、酶学指标的变化。结果:氧化苦参碱组超氧化物歧化酶、谷胱甘肽过氧化物酶活力、一氧化氮水平、诱导型一氧化氮合酶活力显著高于缺血再灌注组,丙二醛显著低于缺血再灌注组(P<0.01)。结论:氧化苦参碱对大鼠缺血再灌损伤的心肌具有保护作用,此作用可能与抑制心肌脂质过氧化、增强抗氧化能力有关。 Aim : To observe the effect of oxymatrine (OMT) on myocardial ischemic reperfusion (IR) injury in the rat heart in vitro, Methods:Forty-five adult Wistar rats were randomly allocated into three groups: normal control groups, IR group, and OMT group. The myocardial ischemia was induced by coronary artery ligation for 30 min and reperfusion for 60 min. Myocardial NO, malondialdehyde (MDA) , the activities of NOS, superoxide dismutase (SOD) , and glutathione peroxidase(GSH-PX) were determined. Results: Compared with IR group, the levels of myocardial SOD, GSH-PX, NO, and NOS were significantly higher while the level of MDA was lower in OMT group ( P 〈 0.01 ). Conclusion : OMT plays a protective role in rat myocardial IR injury in vitro which may involve in its inhibiting lipid peroxidation and enhancing anti-oxidation.
