摘要
目的:观察开口箭皂苷对甲状腺髓样癌TT细胞(MTC-TT)及荷瘤小鼠模型中SUMO特异性蛋白酶2(SENP2)的水平变化,探讨开口箭皂苷治疗甲状腺髓样癌的作用机制。方法:不同浓度的开口箭皂苷作用于MTC-TT细胞,分别给予2、4、8、16、32、64μg/m L开口箭皂苷处理24、48、72 h,采用CCK8法检测各浓度下MTCTT细胞增殖抑制率并计算出半数抑制浓度(IC50),将实验分为4组,低浓度组(TL组,浓度为0.5 IC50开口箭皂苷)、中浓度组(TM组,浓度为IC50开口箭皂苷)、高浓度组(TH组,浓度为2 IC50开口箭皂苷)和对照组(N组,培养基中无开口箭皂苷)。将药物作用于各浓度组MTC-TT细胞48 h后,流式细胞术检测细胞凋亡;Western blot检测MTC-TT细胞中SENP2的蛋白表达;RT-PCR检测MTC-TT细胞中SENP2的基因表达。结果:开口箭皂苷对MTC-TT细胞增殖有抑制作用(P〈0.05),且随浓度增加抗增殖作用越强(P〈0.05或P〈0.01),开口箭皂苷对MTCTT细胞的IC50值为31.54μg/m L;TM组和TH组G0/G1期细胞相对减少(P〈0.05或P〈0.01),TL、TM、TH组G2/M期细胞增加(P〈0.05或P〈0.01),细胞阻滞在G2/M期;TT细胞株中SENP2的蛋白和m RNA表达水平均显著下降。结论:开口箭皂苷可以抑制MTC-TT细胞增殖和诱导其凋亡,其作用机制可能与抑制SENP2的表达有关。
Objective:To observe the effect of total saponins of Tupistra chinensis(TST)on expression of SUMO specific protease 2(SENP2)in the medullary thyroid carcinoma TT(MTC-TT)cells and tumor bearing mice,and explore the mechanism of TST in treating MTC. Methods:Treating TT cells with TST at 24,48,72 h and different concentration(2,4,8,16,32,64 μg/m L)respectively,the inhibitory rate of MTC-TT cells was measured by CCK8 and the inhibitory concentration(IC50)was calculated. The experiment was divided into four groups:low concentration group(TL group)with 0.5 IC50 TST,medium concentration group(TM group)with IC50 TST,high concentration group(TH group)with 2 IC50 TST,the control group(N group)without TST. After the cells had been treated with different concentration of TST for 48 h,the cell apoptosis was detected by flow cytometry. RT-PCR and Western blot were used to detect the m RNA and protein expression of SENP2 in MTC-TT cells and tumor tissues. Results:TST had the inhibitory action on the proliferation of MTC-TT cells(P〈0.05),and the stronger the anti proliferation effect with the increase of concentration(P〈0.05 or P〈0.01),the IC50 value of TST to MTC-TT cells was 31.54 μg/m L,and cells in TM and TH groups decreased in G0/G1(P〈0.05 or P〈0.01),cells in TL,TM and TH groups decreased in G2/M(P〈0.05 or P〈0.01),arrested cells at G2/M phase;and the expression levels of SENP2 and m RNA in TT cell lines were significantly decreased. Conclusion:TST could inhibit MTC-TT cell proliferation and induce apoptosis,which may be related to the inhibition of SENP2 in MTC-TT cells.
