响应面法优化提取高粘度ι-卡拉胶

Optimizing the Extraction of High Viscosity ι-Carrageenan via Response Surface Method

摘要 针对目前ι-卡拉胶提工艺水平不高、软凝胶性不佳的问题,采用水提醇沉法制备ι-卡拉胶,根据中心组合试验设计原理,应用响应面法对碱处理及煮胶环节进行联合优化。优化的工艺参数为KOH碱处理温度46.5℃、煮胶温度93℃、煮胶时间3 h,在此条件下ι-卡拉胶得率为31.76%,凝胶强度为82.56 g/cm^2,粘度为212.52 mPa·s,粘度明显高于市售产品的62.2 mPa·s。结果表明,该优化工艺合理,ι-卡拉胶得率高、软凝胶特性好。 Aiming to resolve the problems of lower extraction technology level and bad viscosity property forι-carrageenan, water-extracted alcohol precipitation method is used to prepare ι-carrageenan. According to the center combination test design, response surface method is used for alkali treatment and scouring. The optimized parameters are 46.5℃ of KOH alkali treatment. The temperature is 93℃ and the consumed time is 3 h of cooking sage. Under this condition, the yield is 31.76 %, the gel strength is 82.56 g/cm^2 and the viscosity is 212.52 mPa·s respectively. The viscosity is obviously superior to commercial products with 62.2 mPa·s. The extraction process is reasonable for high yield and nice soft gel properties.
