海带多糖的分离及其体外抗氧化活性的探究 被引量:7

摘要 采用大孔树脂法分离海带多糖,并对海带多糖的体外抗氧化进行初步评价。以HZ801、HZ816、HP20、HP100、D3520五种大孔树脂为试样,探究最适分离海带多糖的树脂型号。结果表明,大孔树脂D3520对海带多糖的分离效果最好,其最适吸附条件为流速2BV·h-1、pH 5,吸附率为71.14%;洗脱的最适条件为洗脱剂乙醇体积分数45%、流速2BV·h-1,洗脱率为86.27%。药效学以抗氧化消除·OH自由基的消除率为指标,VC为阳性对照,结果表明海带多糖对·OH的有较明显消除效果,当浓度为2.0mg/mL时,清除率为38.36%,说明海带多糖具有一定抗氧化活性。 Macroporous resin method was used to extract laminaria polysaccharide sulphate from laminaria Japonica. The paper evaluated the antioxidant of laminaria in terms of pharmacodynamic. Five macroporous resin methods such as HZ801, HZ816, HP20, HP100, D3520 were tested to explore the best resin model and extraction conditions. The results showed that D3520 worked best and its most suitable adsorption condition was at a flow rate of 2 BV·h^-1. The concentration was pH 5. The adsorption rate was 71.14 %. The optimal desorption was 45 % of ethanol concentration at a flow rate of 2 BV·h^-1. The desorption rate was 86.27 %. The pharmacodynamic took elimination rate of antioxidant eliminating ·OH radicals as a mark and VC as positive control. It was proven that laminaria polysaccharide sulphat had positive effect on eliminating·OH radicals. When the viscosity was 2.0 mg/mL, the elimination rate was 38.36 %. It explained the antioxidantion of laminaria polysaccharide sulphate.
