摘要
目的分析影响流行性感冒病毒(流感病毒)细胞培养阳性的相关因素。方法对2014年4月—2017年3月采集的流感样病例咽拭子标本进行PCR核酸检测,对检测阳性的样本进行细胞分离培养,以分离得到的阳性样本为病例组,阴性样本为对照组,进行对照分析研究,用Logistic回归对影响病毒分离阳性的因素进行分析。结果1 298份PCR核酸检测阳性样本中,分离阳性率为22.80%(296/1 298)。其中,季节性H3N2占13.79%(179/1 298),甲型流感病毒H1N1占1.54%(20/1 298),乙型流感病毒(Victoria)占7.24%(94/1 298),乙型流感病毒(Yamagata)占0.23%(3/1 298)。样本来源、发病采样时间间隔、原始样本PCR核酸检测结果、不同采样机构和采样至收样时间间隔对病毒分离结果差异均有统计学意义(P<0.05)。结论缩短收样至接种时间间隔以及提高医疗机构的采样水平是提高流感病毒分离阳性率的关键。
Objective To analyze the related influential factors for the positive influenza virus by cell culture. Methods The throat swab specimens were collected from ILI cases from April, 2014 to March, 2017 in Urumqi, and then PCR positive specimens were cultured for isolation of influenza virus by MDCK cell culture. The specimens with positive influenza virus isolates as case group and those negative specimens as control group were involved for case-control study. Logistic regression was used in analysis of the related influential factors of isolated positive influenza virus. Results The isolated positive rate was 22.80% (296/1 298) in 1 298 P CR positive specimens, of which seasonal H3N2 accounted for 13.79% (179/1 298) , fluenza A virus (HIN1) 1.54% (20/1 298) , influenza B virus (Victoria) 7.24% (94/1 298) and influenza B virus (Yamagata) 0.23% (3/1 298). Interval between sampling and sample receiving and the different sampling mechanism were identified as the influential factors of positive influenza virus ( P〈0.05 ). Conclusion The two cxucial factors were involved to improve the positive rate of influenza virus isolation, including shortening the interval between sampiing and receiving and improving sampling level.
