动物肝脏中维生素A的反相高效液相色谱快速测定法 被引量:1

摘要 目的建立1种快速测定动物肝脏中维生素A含量的反相高效液相色谱法。方法样品经无水乙醚提取,采用Acclaim 120 C18色谱柱(4.6×250 mm)分离,流动相为甲醇-水(95∶5,V/V),紫外检测波长为300 nm。结果维生素A在试验条件下能完全分离,其线性范围为2~50μg/ml,检出限为0.1μg/ml。维生素A在鸡肝、鸭肝和猪肝中的添加水平为1.0 mg/100 g,鸡肝、鸭肝和猪肝中维生素A的含量在0.72~1.93 mg/100 g,添加回收率为92.49%~94.04%,相对标准偏差(RSD)为0.85%~3.06%。结论该方法操作简便,能测定动物肝脏中维生素A的含量,精密度和回收率高,重复性好,符合食品检测国家标准相关要求。 [Objective] To establish a method for rapid determination of vitamin A in animal liver by reversed phase high performance liquid chromatography.[Methods]The samples were extracted with anhydrous ether and separated by using Acclaim120 C18 column(4.6×250 mm). Methanol-water(95∶5,V/V) was the mobile phase,and the UV detection wavelength was 300 nm.[Results]Vitamin A could be separated completely under the experimental conditions,the linear range was 2-50 μg/ml,and the detection limit was 0.1 μg/ml.The add level of vitamin A in chicken liver,duck liver and pork liver was 1.0 mg/100 g,and the vitamin A contents in chicken liver,duck liver and pork liver were 0.72-1.93 mg/100 g. The recoveries were 92.49%-94.04%,and the relative standard deviation(RSD) were 0.85%-3.06%.[Conclusion]This method is simple and convenient. It can be used to determine the content of vitamin A in animal liver with high precision,high recovery rate and good repeatability. The method meets the requirements of national standards for food testing.
