摘要 分析高效液相色谱法测定动物肝脏中维生素A含量的不确定度来源和影响不确定度的因素,为简化评定方法和提高检测质量提供依据。根据JJF 1135-2005《化学分析测量不确定度评定》和JJF 1059.1-2012《测量不确定度评定与表示》中的有关规定,建立测定动物肝脏中维生素A含量的不确定度的数学模型,逐层对不确定度来源进行分析。由各不确定度分量评定结果可以看出,高效液相色谱法测定动物肝脏中维生素A的含量:当鸭肝中维生素A的含量为0.73 mg/100 g时,其扩展不确定度为0.04 mg/100 g(k=2);当猪肝中维生素A的含量为1.12 mg/100 g时,其扩展不确定度为0.06 mg/100 g(k=2);当鸡肝中维生素A的含量为1.97 mg/100 g时,其扩展不确定度为0.12 mg/100 g(k=2)。高效液相色谱法测定动物肝脏中维生素A含量的不确定度主要由重复测定、标准溶液配制、样品测定和校准曲线拟合等因素引入。 To analyze the sources and influencing factors of content uncertainty of vitamin A in animal liver by highperformanceliquidchromatography(HPLC),soastosimplifytheevaluationmethodandincreasethetesting quality.According to JJF 1135-2005 Evaluation of Uncertainty in Chemical Analysis Measurement and JJF 1059.1-2012 Evaluation and Expression of Uncertainty in Measurement,a mathematical model for the determination of vitamin A content in animal liver was established,and the source of uncertainty was analyzed by layer by layer.The results of the uncertainty components showed that the content of vitamin A in animal liver was determined by HPLC:an expanded uncertainty of 0.04 mg/100 g(k=2)was obtained for the determination of the presence of 0.73 mg/100 g Vitamin A in duck liver,an expanded uncertainty of 0.06 mg/100 g(k=2)was obtained for the determination of the presence of 1.12 mg/100 g Vitamin A in pig liver,an expanded uncertainty of 0.12 mg/100 g(k=2)was obtained for the determination of the presence of 1.97 mg/100 g Vitamin A in chicken liver.The main sources of uncertainty of vitamin A in animal liver by HPLC were repetitive measurement,preparing the standard solution,sample determination,and curve fitting.
