IL-2对布鲁菌侵染小鼠巨噬细胞释放细胞因子及吞噬能力的调节作用 被引量:8

摘要 为研究白介素2(IL-2)对被布鲁菌M5株侵染的巨噬细胞(MΦ)所释放的细胞因子(TNF-α、IFN-γ和NO)及吞噬能力的调节作用,通过对M5侵染的MΦ添加不同剂量的IL-2,用ELISA试剂盒检测MΦ(侵染的和未侵染的)在不同时间点所释放的细胞因子TNF-α、IFN-γ和NO含量的变化;用CFU涂板法计算胞内活菌数,从而计算巨噬细胞的吞噬指数,以此来探讨IL-2对MΦ在被M5侵染后的免疫调节作用。结果表明,IL-2作用于被M5侵染的MΦ所释放的细胞因子(TNF-α、IFN-γ和NO)的含量与对照组相比有显著的提高(P<0.05),且存在一定的含量和时间的依赖性;IL-2对巨噬细胞吞噬M5的能力在一定程度上有促进作用,随着IL-2含量的增加,胞内活菌数亦增加,存在浓度依赖性。通过试验结果可知,IL-2可以促进MΦ活化,促进MΦ分泌TNF-α、IFN-γ和NO,增强机体免疫应答,为临床应用IL-2研究及治疗布鲁菌病提供新的思路和理论依据。 To investigate the regulatory effect of IL-2 on the release of cytokines (TNF-α, IFN-γ and NO) and phagocytic capacity of the macrophages infected with Brucella M5 strain,different doses of IL-2 were added to infected MФ , and the content changes of TNF-α, IFN-γ and NO released by MФ (infection and no infection) infected with M5 at different time points were detected with ELISA kit; the number of intracel- lular viable bacteria and the phagocytic index of macrophages were calculated by CFU coating method in order to explore the effect of IL-2 on the immune regulation of MФ after M5 infection. The test results showed that the cytokines (TNF-α, IFN-γ and NO) were released by M5 infected MФ, and the levels of cytokines were significantly increased compared with the control group (P〈0.05) under the regulation of IL-2, and there was a certain concentration and time dependence; IL-2 could promote the phagocytosis of macrophages to a certain extent, and the number of intracellular viable bacteria was also increased with the increase of IL-2 concentration, which was concentration-dependent. The results of the present study showed that IL-2 can promote the activation of MФ and the secretion of TNF-α, IFN-γ and NO by MФ and enhance the immune response of the body. It may provide a new idea and theoretical basis for the clinical use of IL-2 and treatment of brucellosis.
