Expression of micro RNA-92a in diffuse large B cell lymphoma and its mechanism of action

摘要 目的分析miR-92a在弥漫大B细胞淋巴瘤(DLBCL)患者中的表达与临床病理特征的关系,观察miR-92a对弥漫大B淋巴瘤细胞系OCI-LY10增殖、侵袭能力的影响。方法采用实时荧光定量PCR方法检测86例DLBCL患者肿瘤组织和22例正常淋巴结组织中miR-92a的表达,分析miR-92a表达水平与患者临床病理特征的关系。对弥漫大B淋巴瘤细胞系OCI-LYl0进行miR-92a反义核苷酸(ASO)转染并培养,MTT法检测细胞增殖能力,Transwell小室检测细胞侵袭能力,real-time PCR方法与Western blot方法检测细转染miR-92a ASO后各组细胞KLF4的m RNA和蛋白的表达。结果DLBCL患者淋巴结组织中miR-92a表达水平显著高于对照组(P<0.05)。miR-92a在DLBCL患者中的表达与疾病分期显著相关(P<0.05),但与患者年龄、性别无关(P>0.05)。与对照组相比,miR-92a ASO组OCI-LYl0细胞的增殖侵袭能力明显降低,KLF4蛋白与m RNA表达水平显著升高(P<0.05)。结论miR-92a在DLBCL中表达上调,其促进OCI-LY10细胞增殖侵袭的作用机制可能与下调KLF4蛋白表达有关。 Objective To investigate the relationship between the expression of miR-92a and clinicopathologic characteristics in diffuse large B cell lymphoma (DLBCL), and the effect of miR-92a on proliferation and invasion of diffuse large B lymphoma cell line OCI-LYIO. Methods The level of miR-92a was detected by real-time PCR in 86 cases of DLBCL tissues without invasion of tumor cells and 22 cases of normal lymph node tissues, and then the relationship between miR-92a expression and elinicopathologic characteristics was analyzed. The miR-92a antisense oligonueleotides(ASO) were transfeeted into diffuse large B cell lymphoma cell line OCI-LYIO, the proliferation and invasion of OCI-LY10 cells were evaluated by MTT assay and Transwell assay, the expression level of KLF4 in OCI-LY10 cells was detect by real-time PCR and Western blot. Results The expression level of miR-92a was significantly higher in lymph node tissue of DLBCL patients than in the control group(P〈0.05), and related to the stage of disease (P〈0.05), but not to the age and sex of patients(P〉0.05). The proliferation and invasion ability of OCI-LY10 cells were significantly decreased in miR-92a ASO group than in the control group, lout the expression levels of KLF4 protein and mRNA were significantly increased(P〈0.05). Conclusion The promoting proliferation and invasion of OCI-LY10 cells by miR-92a might be related to down-regulating the expression of KLF4 protein.
