当归补血汤对内质网应激特有Caspase-12凋亡途径影响 被引量:9

摘要 目的:研究当归补血汤对内质网应激特有的Caspase-12凋亡途径的影响,探讨当归补血汤对肾脏的保护机制。方法:60只SD大鼠腹腔内注射链脲佐菌素(STZ)结合高脂高糖饮食建立大鼠糖尿病肾病模型(DN),喂养8周后,观察大鼠的毛色、精神状态等一般状态;酶偶联比色法检测尿蛋白、BUN、尿肌酐等反应肾功能的相关指标;光镜观察其肾脏病理变化;RT-PCR、western Blot及免疫组化检测GRP78、Caspase-12的蛋白及m RNA表达水平;TUNEL检测肾组织细胞凋亡情况。结果:DN组大鼠空腹血糖、尿蛋白、BUN等明显升高,肾功能受损严重,肾脏组织GRP78、Caspase-12的蛋白及m RNA含量增加,光镜下肾脏结构病变明显,肾组织细胞凋亡率增高;当归补血汤治疗后肾功能损伤减轻,GRP78、Caspase-12的蛋白及m RNA含量降低,光镜下肾组织病变改善,大鼠肾组织凋亡率降低。结论:当归补血汤能抑制内质网应激特有Caspase-12凋亡途径,改善糖尿病肾病大鼠肾功能,减少肾组织过度凋亡。 Objective:To study the effect of Danggui Buxue Decoction on endoplasmic reticulum stressspecific Caspase-12 apoptosis way in diabetic rats and to explore its protection mechanisms. Methods:Sixty SD rats were given STZ + high-fat feed,sacrificed after eight weeks,observed the general condition of these rats,measured blood glucose,and the indicators that related with renal function,enzyme-linked immunosorbent assay to detect the contents of urinary MAU,optical microscope were used to observe the kidney structure change. The total expression of GRP78/Caspase-12 m RNA and protein was determined by real-time PCR,Western blotting and immunohistochemical respectively. The apoptosis of kidney tissues cells was determined by TUNEL assay. Results:The DM group had a significantly higher level of blood glucose,MAU,BUN,serumcreatinine. renal function is seriously damaged;The expression of GRP78 and Caspase-12,both m RNA and protein,were higher;optical microscopically kidney damage was significantly serious;The amount of kidney tissues apoptosis cells significantly were higher. After treatment with Danggui Buxue Decoction treatment,renal function was relieved;The expression of GRP78 and Caspase-12,both m RNA and protein,were dropped;The amount of kidney tissues apoptosis cells were down;optical microscopically kidney damage was significantly relieved. Conclusion:This study proved that Danggui Buxue Decoction can inhibite endoplasmic reticulum stress-specific Caspase-12 apoptosis way,protects the renal function,decline kidney tissues apoptosis in diabetic rats.
