摘要
【目的】探讨刀豆蛋白A(Con A)诱导急性免疫性肝损伤Wistar大鼠模型的剂量和注射途径。【方法】(1)按不同剂量Con A尾静脉注射分组:将42只雌性Wistar大鼠随机分为A、B、C、D、E、N组,每组7只。N组为空白对照组,大鼠尾静脉注射生理盐水。A、B、C、D、E组大鼠尾静脉注射Con A,剂量分别为4、8、16、30、40 mg/kg。注射后第8 h,检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、白蛋白(ALB)及细胞因子白细胞介素(IL)-2、IL-10、干扰素(IFN)-γ、肿瘤坏死因子(TNF)-α的表达水平,采用苏木素—伊红(HE)染色法观察肝组织病理变化。(2)按Con A不同注射途径分组:将21只Wistar大鼠随机分为正常对照组、腹腔注射组、尾静脉注射组,每组7只,尾静脉注射组和腹腔注射组分别经尾静脉和腹腔注射16 mg/kg Con A。注射后第8 h,检测血清ALT、AST、ALB水平。【结果】实验结束时Con A各剂量组大鼠非正常死亡数:A、B、C组0只,D组2只,E组7只。A、B组的肝组织可见少量点状坏死,炎性细胞浸润,小叶结构基本完整;C、D组可见明显的桥接样坏死。尾静脉注射组大鼠血清ALT、AST、ALB水平显著升高,与正常对照组比较,差异有统计学意义(P<0.05)。【结论】尾静脉注射16 mg/kg Con A可成功诱导Wistar大鼠急性免疫性肝损伤模型。
Objective To explore the dosage and injection method of concanavalin A(Con A)for inducing Wistar rats into the acute hepatic injury model. Methods(1)According to the dosage of Con A,42 Wistar rats were randomly divided into groups A,B,C,D,E,N,7 rats in each group. Group N was given tail intravenous injection of normal saline as normal control group. Groups A,B,C,D,E were given intravenous injection of4, 8, 16, 30, 40 mg/kg of Con A respectively. At the 8 th hour after modeling, the levels of alanine transaminase(ALT),aspartate aminotransferase(AST),albumin(ALB),interleukin(IL)-2,IL-10,interferon(IFN)-γ,and tumor necrosis factor(TNF)-α were detected. And HE staining was used to observe the pathological feature of hepatic tissue.(2)According to the injection method of Con A,21 Wistar rats were randomly divided into normal control group, intraperitoneal injection group and tail intravenous injection group, 7 rats in each group. The dosage of Con A for the rats in intraperitoneal injection group and tail intravenous injection group was16 mg/kg. At the 8 th hour after modeling,the levels of serum ALT,AST,and ALB were determined. Results The number of abnormal deaths in various dose Con A groups at the end of each experiment was 0 in groups A,B,C,and 2 in group D,and 7 in group E. A small amount of spotty necrosis,inflammatory cell infiltration,and hepatic lobule with almost integrity of structure were found in groups A, B, while obvious bridging-like necrosis was seen in groups C,D. Serum ALT,AST,and ALB levels in intraperitoneal injection group had no statistically significant difference as compared with the normal control group. Conclusion Tail intravenous injection of 16 mg/kg of Con A can be used to induce an acute immunological liver injury rat model successfully.
