摘要
检测癌胚抗原相关黏附分子1(CEACAM1)和基金属蛋白酶-2(MMP-2)在AGS和SGC-7901胃癌细胞系中的表达,探讨在胃癌细胞发生发展和侵袭转移中的作用。采用实时定量PCR检测两种细胞株中CEACAM1和MMP-2的mRNA表达;采用Western blot检测两种细胞株中CEACAM1和MMP-2的蛋白相对表达量;采用Trasnwell小室分析两种细胞株的侵袭能力。AGS的CEACAM1和MMP-2的mRNA及蛋白表达量均高于SGC-7901,差异有统计学意义(P<0.05)。AGS的穿膜细胞数为(147.2±3.6),高于SGC-7901的穿膜细胞数(83.5±3.9),差异有统计学意义(P<0.05)。CEACAM1和MMP-2的表达与胃癌细胞分化程度相关,且表达程度越高,其侵袭、转移能力越强。
To investigate the expression of CEACAM1 and MMP-2 and to explore the effects of CEACAM1 and MMP-2 on invasion and metastasis of gastric cancer cell lines AGS and SGC-7901,RT-PCR was used to detect the expressions of mRNA in CEACAM1 and MMP-2. Western blot was used to measure the relative expression of proteins in CEACAM1 and MMP-2. Trans well chambers were used to investigate the migration of the two cell lines. The mRNA level and relative protein content in AGS were higher than those in SGC-7901(P〈0. 05). For AGS,the count of transmembrane cell was more than SGC-7901(147. 2 ± 3. 6 vs. 83. 5 ± 3. 9,P〈0. 05). The expression of CEACAM1 and MMP-2 was connected with the differentiation degree of gastric cancer cell,and higher levels may promote the invasion and metastasis of cancer cell.
