摘要
【目的】为了明确四川脆红李品种上新近爆发的一种未知病害,解除脆红李大面积发生的"火烧叶"病害对脆红李产业的威胁。【方法】本文从不同发病地区采集"火烧叶"病叶,通过室内镜检,病原分离、纯化、致病性测定以及病原菌再分离鉴定等方法,对引起脆红李"火烧叶"病害的病原进行了形态学和分子生物学鉴定。【结果】引起四川省多个脆红李产区的"火烧叶"病原为链格孢菌[Alternaria alternata(Fr.)Keissler]。【意义】脆红李"火烧叶"病病原鉴定为"火烧叶"病害发生流行规律的深入研究和进一步制定科学的防治措施奠定了基础。
【Objective】The objective of this study is to identify the pathogen causing‘flaming leaves disease'on plum in Sichuan,and to provide a basis for disease prevention.【Method】Diseased samples from different areas were collected. Pieces of infected tissue were surface disinfested and plated on acidified potato dextrose agar( PDA). The growing edges of any fungal hyphae developing from the tissues were then transferred aseptically to another PDA plate. Identification of the pathogenic fungus was carried out according to the characteristics of fungal morphology,pathogenicity,partial r DNA-ITS and polygalacturonase gene sequence analysis.【Result】Based on the study of morphology,cultural feature,pathogenicity testing and partial sequences of r DNA-ITS and polygalacturonase gene,the pathogenic of plum‘flaming leaves disease'is Alternaria alternata( Fr. : Fr.) Keissler. 【Conclusion】The clarification of pathogen would benefit the study on epidemiology and control of this disease.
