神秘果树叶提取物降尿酸作用及其有效成分鉴定 被引量:7

Decreasing Uric Acid Effect of the Extracts of Miracle Fruit(Synsepalum dulcificum)' Leaf and Identification of Its Active Ingredient

摘要 黄嘌呤氧化酶是嘌呤代谢生成尿酸过程中的关键酶,对于高尿酸血症的发生和发展起至关重要的作用。本文构建了氧嗪酸钾诱致高尿酸血症大鼠模型,对比研究了神秘果树叶乙醇、水提取物在动物体内的降尿酸作用,采用现代色谱、波谱技术导向分离具有黄嘌呤氧化酶抑制活性的化合物。研究结果表明:神秘果树叶乙醇、水提取物在动物体内均具有一定的降尿酸作用。乙醇提取物降尿酸活性强于水提取物,并可显著增加高尿酸血症大鼠血清肌酐含量及降低尿素氮含量,呈现较好的护肾活性。从神秘果树叶中分离鉴定出3种槲皮素类化合物,包括槲皮素、槲皮素-3-O-α-L-鼠李糖苷以及金丝桃苷。3种槲皮素类化合物在乙醇提取物中的含量从高到低依次为:槲皮素-3-O-α-L-鼠李糖苷【1.74 g/(100 g)】、金丝桃苷【0.77 g/(100 g)】、槲皮素【0.17 g/(100 g)】。黄嘌呤氧化酶抑制活性从强到弱依次为:槲皮素、金丝桃苷、槲皮素-3-O-α-L-鼠李糖苷。神秘果树叶提取物以及槲皮素类化合物可用作保健食品功效因子,用以预防与治疗高尿酸血症。 Xanthine oxidase is a hepatic enzyme and catalyzes two-step oxidation of hypoxanthine to xanthine and then xanthine to uric acid, which plays an important role in the occurrence and development of hyperuricemia. For the hyperuricemia study, potassium oxonate was used to induce acute hyperuricemia. The study aimed to explore the potential of the miracle fruit leaf ethanol and water extracts to prevent and treat hyperuricemia. On the basis of in vitro xanthine oxidase inhibitory(XOI) activity-guided purification, bioactive compounds were purified and identified using modern chromatography and spectroscopy. The miracle fruit leaf ethanol and water extractscould reduce the serum uric acid levels of hyperuricemic rats and do not exert any adverse effect on rat body weight. The miracle fruit leaf ethanol extract was more effective than water extract and significantly increased the serum creatinine and reduced the urea nitrogen of oxonate-treated rats, indicating that it possessed the renal protective effect. It is the first time that quercetin, quercetin-3-O-α-L-rhamnopyranoside and hyperoside have been identified in the miracle fruit leaf. The amounts of three compounds in the miracle fruit leaf extract were in a decreasing sequence: quercetin-3-O-α-L-rhamnopyranoside [1.74 g/(100 g)]〉 hyperoside [0.77 g/(100 g)] 〉quercetin [0.17 g/(100 g)]. In vitro XOI activities were in the order of quercetin〉 hyperoside quercetin-3-O-α-L-rhamnopyranoside. The miracle fruit leaf extract, together with quercetin derivatives could be useful agents for prevention and treatment of hyperuricemia and be effectively used for pharmaceutical and nutraceutical applications.
