摘要
目的:确定一种可以快速富集生乳中沙门氏菌的方法条件,可用于生乳沙门氏菌的快速检测。方法:通过对缓冲液种类及浓度,去除脂肪、蛋白质等干扰物质方式,洗脱液种类及浓度的研究,确定生乳快速富集沙门氏菌条件,并将该方法与国标方法进行对比验证实验,确定其准确性和灵敏性。结果:缓冲液选择0.01 mol/L PBS,采用滤纸过滤、离心和0.22μm微孔滤膜过滤相结合的方法去除脂肪和大分子蛋白质,洗脱液选择5 mL PBS+5 mL TTB混合液,沙门氏菌的富集分离效果最佳。经实验验证,按照该方法检测灵敏度可达到10 cfu/mL,高于国标方法。结论:建立了最佳的快速富集生乳中沙门氏菌的方法条件,使沙门氏菌检测时间从(4~7)d缩短至50 min,敏感性和特异性较好。
Objective: A method for rapid enrichment of Salmonella in raw milk was confirmed, which could be used for rapid detection of Salmonella in raw milk. Method: Through the research of buffer type and concentration, the removal of fat, protein and other interfering substances, the elution liquid type and concentration, to determine the conditions for rapid enrichment of Salmonella in raw milk. The method was compared with the national standard method to verify the accuracy and sensitivity. Result: When the buffer was selected 0.01 mol/L PBS, the fat and macromolecular protein were removed by filter paper filtration, centrifugation and 0.22 μm microporous membrane filtration, the elution solution was selected 5 m L PBS+5 m L TTB mixed liquid, the enrichment and separation effect of Salmonella was the best. The test results showed that the sensitivity of this method could reach 10 cfu/m L, which higher than the national standard method. Conclusion: The optimal method for rapid enrichment of Salmonella in raw milk was established, which could shorten the detection time of Salmonella from(4~7)d to 50 min, and the sensitivity and specificity were better.
