外周血中Let-7e、微小RNA-206、微小RNA-605及微小RNA-1236表达与原发性高血压的相关性研究 被引量:3

摘要 目的:探讨外周血中Let-7e、微小RNA-206、微小RNA-605及微小RNA-1236与原发性高血压的相关性。方法:选取2014-07到2016-12在内蒙古自治区医科大学附属医院收治的169例原发性高血压患者(原发性高血压组)及同期体检健康者143例(正常对照组),应用血压计测定研究对象血压,通过反转录实时定量聚合酶链式反应(PCR)测定Let-7e、微小RNA-206、微小RNA-605及微小RNA-1236在外周血中的表达量。并进行微小RNA表达量与高血压发病相关性及原发性高血压发病的多因素Logistic回归分析。结果:原发性高血压组Let-7e、微小RNA-605的表达量均高于正常对照组,分别为1.42±0.15 vs 1.00±0.21(P<0.05)和2.14±0.22 vs 1.00±0.11(P<0.05);而原发性高血压组中微小RNA-206、微小RNA-1236的表达量均低于正常对照组,分别为0.69±0.12 vs 1.00±0.18(P<0.05)和0.46±0.17 vs 1.00±0.31(P<0.05),差异均有统计学意义。相关性分析显示,Let-7e的表达量与收缩压呈显著正相关(r=0.562,P<0.05);微小R-206表达量与收缩压呈显著负相关(r=-0.582,P<0.05),与舒张压呈显著正相关(r=0.507,P<0.05);微小RNA-605的表达量与收缩压呈显著正相关(r=0.625,P<0.05);微小RNA-1236的表达量与舒张压呈显著负相关(r=-0.598,P<0.05),与体重指数呈显著正相关(r=0.690,P<0.05)。多因素Logistic回归分析显示,体重指数的升高显著增加患原发性高血压的危险(95%CI:1.7933~3.9941,P<0.01),Let-7e和微小RNA-605的表达量的升高显著增加患高血压的危险(95%CI:1.5378~1.7632,P<0.05和95%CI:1.5983~2.4203,P<0.01),而微小RNA-206和微小RNA-1236的表达量的降低,也显著增加原发性高血压患病的风险(95%CI:0.5546~0.8782,P<0.01和95%CI:0.2876~0.5614,P<0.01)。结论:外周血中Let-7e、微小RNA-206、微小RNA-605及微小RNA-1236与原发性高血压的发病有密切的关系。 Objective: To explore the relationship between peripheral blood levels of Let-7 e, miR NA-206, miR NA-605, miR NA-1236 and essential hypertension(EH) occurrence. Methods: Our research included in 2 groups: HE group, n=169 patients treated in our hospital from 2014-07 to 2016-12 and Control group, n=143 healthy subjects from physical examination at the same period. Blood pressure was measured by sphygmomanometer, the expression levels of peripheral Let-7 e, miR NA-206, miR NA-605 and miR NA-1236 were detected by reverse transcription quantitative polymerase chain reaction(RT-qP CR). Relationship between miR NA levels and HE occurrence was studied by multivariate Logistic regression analysis. Results: Compared with Control group, EH group showed increased expressions of Let-7 e and miR NA-605(1.42±0.15) vs(1.00±0.21) and(2.14±0.22) vs(1.00±0.11), both P〈0.05; decreased expressions of miR NA-206 and miR NA-1236(0.69±0.12) vs(1.00±0.18) and(0.46±0.17) vs(1.00±0.31), both P〈0.05. Correlation analysis presented that peripheral level of Let-7 e was positively related to systolic blood pressure(r=0.562, P〈0.05); miR NA-206 was negatively to systolic blood pressure(r=-0.582,P〈0.05) and positively related to diastolic blood pressure(r=0.507, P〈0.05); miR NA-605 was positively related to systolic blood pressure(r=0.625, P〈0.05); miR NA-1236 was negatively related to diastolic blood pressure(r=-0.598, P〈0.05) and positively related to BMI(r=0.690, P〈0.05). Multivariate Logistic regression analysis indicated that elevated BMI could increase the risk of EH occurrence(OR=2.6412, 95%CI 1.7933-3.9941, P〈0.01), elevated expression of Let-7 e and miR NA-605 could increase the risk of EH occurrence(OR=1.6505, 95%CI 1.5378-1.7632, P〈0.05) and(OR=1.9893, 95%CI 1.5983-2.4203, P〈0.01); reduced expression of miR NA-206 and miR NA-1236 could also increase the risk of EH occurrence(OR=0.7164, 95%CI 0.5546-0.8782, P〈0.01)
