摘要
目的研究survivin及caspase-3在分化型癌组织中的阳性表达及两者的相互关系。方法选取西安市中心医院2010年1月~2016年10月间分化型甲状腺癌石蜡包埋标本56例。应用免疫组织化学法(SP法),检测survivin、caspases-3在分化型甲状腺癌中的表达。结果结节性甲状腺肿组、甲状腺癌旁组survivin蛋白的表达差异无统计学意义(P>0.05);结节性甲状腺肿组survivin表达与DTC组差异有统计学意义(P<0.05);结节性甲状腺肿组、甲状腺癌旁组caspase-3蛋白的表达经检验两者间差异无统计学意义(P>0.05);结节性甲状腺肿组caspase-3蛋白表达与DTC组(30/56,53.5%)差异有统计学意义(P<0.05);survivin和caspase-3在DTCⅠ~Ⅱ期的阳性表达率与DTCⅢ~Ⅳ期的阳性表达率及有无淋巴结转移组中比较,差异有统计学意义(P<0.05);caspase-3差异无统计学意义(P>0.05)。survivin表达与caspase-3蛋白表达呈负相关(r=-0.325,P<0.01)。结论 survivin与caspase-3介导的细胞凋亡途径参与了甲状腺癌的发生、发展过程。
Objective To evaluate the positive expression and the relationship of Survivin and caspase-3 in differentiated carcinoma. Methods Fifty-six cases of paraffin-embedded specimens of differentiated thyroid carcinoma from January 2010 to October 2016 were selected from Xi＇an Central Hospital. The expression of survivin and caspases-3 in differentiated thyroid carcinoma was detected by immunohistochemistry （SP method）. Results The expression of Survivin protein in nodular goiter group and thyroid carcinoma group was not significantly different （P〉0.05）. The expression of Survivin in nodular goiter group was significantly different from that in DTC group （P〈0.05）. The expression of caspase-3 protein in the nodular goiter group was significantly higher than that in the DTC group （30/56,53.5）, and the expression of caspase-3 protein in the goiter group and thyroid carcinoma group was significantly different （P〈0.05）. The positive expression rate of survivin and caspase-3 in DTC Ⅰ-Ⅱ was significantly different from that in DTC Ⅲ-Ⅳ and lymph node metastasis （P〈0.05）. There was no significant difference in caspase-3. （P〉0.05）. Survivin expression was negatively correlated with caspase-3 protein expression （r=-0.325, P〈0.01）. Conclusion Survivin and caspase-3 are involved in the development of thyroid carcinoma by apoptotic pathway.
