摘要
目的:探讨补肾舒脊颗粒对hBMSC软骨内成骨过程中Wnt/β-catenin信号通路的作用。方法:原代hBMSC传至第3代后分为5组:补肾舒脊颗粒高、中、低剂量组,西药组,空白对照组,进行软骨细胞诱导分化,运用ELISA法检测细胞上清液中DKK-1表达,Western Blot和荧光定量PCR检测SOST、Wnt5a和β-catenin蛋白和基因表达。结果:与空白对照组比较,中药中、高剂量组DKK-1、SOST蛋白水平显著增高(P<0.05,P<0.01);与西药组比较,中药中、高剂量β-catenin蛋白降低(P<0.05)。与空白对照组比较,中药低、中、高剂量组及西药组SOST mRNA水平升高(P<0.05),中药中、高剂量组及西药组Wnt5a mRNA水平下降(P<0.05),中药低、中、高剂量组β-catenin mRNA水平下降(P<0.05);与西药组比较,中药高剂量组SOST mRNA水平升高(P<0.05),β-catenin mRNA水平降低(P<0.05)。结论:补肾舒脊颗粒可能能够抑制hBMSC软骨内成骨过程中的Wnt/β-catenin信号通路。
Objective:To explore the effects of Bushen Shuji Granules(BSG) on Wnt/β-catenin signaling pathway in the process of hBMSC endochondral ossification.Methods:Primary hBMSC were divided into the following five groups after they were passaged to the third generation:Bushen Shugu Granules in high,medium and low dose groups,western medicine control group and blank control group.hBMSC were induced to differentiate into chondrocytes.Enzyme-linked immuno sorbent assay(ELISA) was applied to detect the supernatant DKK-1 expression.Western Blot and fluorogenic quantitative PCR were applied to detect the protein and gene expressions of SOST,Wnt5a and β-catenin.Results:Compared with the blank control group,the DKK1 and SOST protein secretions of the medium and high dose group increased significantly(P〈0.05,P〈0.01); Compared with the western medicine group,the β-catenin protein of the medium and high dose group decreased(P〈0.05).Compared with the blank control group,the levels of SOST mRNA of the low,medium,high dose group and the western medicine group increased(P〈0.05),while the levels of Wnt5 a mRNA of the medium,high dose group and the western medicine group decreased(P〈0.05),and the levels of β-catenin mRNA of the low,medium and high dose group decreased(P〈0.05); Compared to the western medicine group,the level of SOST mRNA of the high dose group increased(P〈0.05),while the β-catenin mRNA decreased(P〈0.05).Conclusion:Bushen Shuji Granules can inhibit the Wnt/β-catenin signaling pathway in the process of hBMSC endochondral ossification.
